Description / Table of Contents: Summary On 50 patients was made a determination of the ocular rigidity before and after an operation for retinal detachement (diathermy and scleralresection) at which the non-operated eye aided as control. It was established that the measures have an influence on the sclera as far as the rigidity of the eyes decreases by the above mentioned measures. The found dates are according to the statistic significant. The decrease of the rigidity conditioned by operation shows no additional effect. There was namely no difference between the eyes of younger and older patients, between the kind of the two performed operations and also no dependence on the grade of refraction of the operated eye.

Notes: Summary The bis-adducts formed by nickel(II) and zinc(II) chelates of 3-cyano-4-aryl-6-phenyl pyridine-2-(1H)thiones with pyridine and N-methylimidazole have been prepared and characterized by microelemental analyses, i.r., u.v., vis., and 1Hn.m.r. spectra and magnetic measurements. Owing to Lewis acid-base interactions, the diamagnetic square planar nickel(II) or zinc(II) chelates interact with two molecules of pyridine or N-methylimidazole in the two axial positions forming the corresponding octahedral bis-adducts.

Notes: Abstract Chickens from Regional Poultry Research Laboratory (RPRL) inbred line 63 are resistant to virally-induced Marek's disease (MD) and lymphoid leukosis (LL) and are relatively strong regressors of virally-induced Rous sarcomas. In contrast, RPRL line 100 chickens are highly susceptible to MD and LL and are weaker regressors of Rous sarcomas than line 63. RPRL lines 100 and 63 differ for alleles at the IgG-1 (G-1) allotype locus, but have identical IgM-1 (M-1) allotype alleles. To test the possible association of the G-1 locus with variations in resistance to virally-induced tumors, homozygous and heterozygous genotypes among F3 crosses were infected. F3 chickens with different G-1 types were comparable in their resistance to MD tumors following inoculation with the JM strain of the MD virus, and for their ability to regress Rous sarcoma tumors induced by the Rous sarcoma virus (RSV) RAV-1. However, following RAV-1 virus infection a smaller proportion of G-1 a /G-1 a F3 or F4 birds developed LL tumors than G-1 a /G-1 e and G-1 e /G-1 e birds. Genes determining immunoglobulin heavy chains were therefore associated with a recessive resistance to B-cell lymphomagenesis in chickens.

Notes: Abstract Time-course studies on the distribution and metabolism of valproate (VPA) in rat brain, liver, and kidney, after intraperitoneal injection of a mixture of [14C]VPA and [3H]VPA, showed that: (1) maximal amount of radioactivity in the various tissues was observed after 30 min from the time the drug was administered; (2) at 30 min the distribution of labeled VPA in brain, liver, and kidney was 17%, 64%, and 19% of the total radioactivity, respectively; (3) at 24 hr more than 97% of the total radioactivity was lost from the tissues and the14C/3H ratios increased significantly with time. Studies on the regional distribution of the drug showed that it is relatively homogeneously distributed. Studies on the subcellular distribution of the drug showed that it is associated mostly with the soluble and mitochondrial fractions, with little radioactivity in the myelin and synaptosomal fractions. Radiochromatography of VPA metabolites in perchloric acid extracts from brain, liver, and kidney revealed the presence of four metabolites. VPA was not incorporated into phospholipids of the neuronal membranes. Furthermore, it had no significant effects on Mg2+-ATPase and (Na++K+)-ATPase in synaptosomes and microsomes obtained either from control or from rats injected with VPA. It was concluded that this antiepileptic drug does not appear to act through its incorporation into neuronal membrane or through its action on the Na+ pump.

Notes: Abstract Studies were conducted on the effects of antiepileptic drugs on the acetylcholine-stimulated32P labeling of phospholipids in rat brain synaptosomes. Of the four antiepileptic drugs investigated in the present study, namely phenytoin, carbamazepine, phenobarbital, and valproate, only phenytoin blocked the acetylcholine-stimulated32P labeling of phosphatidylinositol and phosphatidic acid, and the acetylcholine-stimulated breakdown of polyphosphoinositides. Phenytoin alone, like atropine alone, had no effect on the32P labeling of phospholipids nor on the specific radioactivity of [32P]ATP. Omission of Na+ drastically reduced both the32P labeling of synaptosomal phospholipids and the specific radioactivity of [32P]ATP and furthermore it significantly decreased the phosphoinositide effect. It was concluded that certain antiepileptic drugs, such as phenytoin, could exert their pharmacological actions through their antimuscarinic effects. In addition the finding that phenytoin, which acts to regulate Na+ and Ca2+ permeability of neuronal membranes, also inhibited the phosphoinositide effects in synaptosomes, support the conclusions that Ca2+ and Na+ are probably involved in the molecular mechanism underlying this phenomenon in excitable tissues.

Notes: Summary Metal complexes of the title ligands were characterized in order to determine the factors influencing the stability of chelate isomerism in the same molecule. The ligands were prepared by 1:1 condensation of isonitrosoacetylacetone (Hiso) with eithero-aminophenol (H2 isoaph),p-aminophenol (H2 isopph), or aniline (Hisoanil). The following complexes have been synthesized: [(isoaph)Cu]4, (Hisoaph)2Co, (Hisopph)2 M·nH2O (M=Ni(II), n=2;M=Pd(II), n=0;M=Co(II), n=2), [(isopph) Cu·H2O]2, and (isoanil)2 M (M=Ni(II), Cu(II), Co(II), or Pd(II)). Both chelate rings in these metal complexes are five-membered. Transimination of one −C=N−C6H5 group to −C=NH in (isoanil)2Ni produced a six-membered chelate ring in (isoim)Ni(isoanil). The induced chelate isomerism is ascribed to intermolecular hydrogen bonding of the imino-hydrogen and the basic nitrogen of the same six-membered chelate ring of an adjacent square planar molecule. Other types of hydrogen bondings with the oximato oxygen (intra- or intermolecular) favour the formation of five-membered chelate rings. Analytical, spectroscopic, and magnetic moment data are in accordance with the suggested formulations.
