    Notes: Abstract An intense radiation at 395.0 nm has been observed when lithium vapor is optically pumped in a heat pipe with a pulsed dye laser whose output wavelength is tuned near the Li 2s–4s two-photon resonance transition. The radiation is emitted in the direction along the pump laser beam. It is proposed that the 395.0 nm radiation is mainly generated through three-photon excitation and one-Raman-photon scattering followed by two-cascade (spontaneous) emission. The overall reaction mechanism can be described by a parametric six-wave mixing process. The quantum efficiency of the observed process is estimated to be of the order of 2 × 10−6.

    Notes: The photoabsorption and photoionization cross sections of GeH4 from the ionization threshold to 1060 A(ring) have been measured using synchrotron radiation as the continuum light source. The cross sections for the production of neutral products in the same spectral region have also been determined. Weak diffuse structures were observed in both the photoabsorption and photoionization spectra. The steplike ionization structures in the ionization spectrum were observed. The adiabatic ionization potential was determined to be at 1177.6±0.5 A(ring) which is in excellent agreement with a value recently reported by Ruscic et al. [J. Chem. Phys. 92, 1865 (1990)] using a photoionization mass spectrometry technique. Rydberg series in the 1060–1180 A(ring) region converging to the ground state of GeH4+ have been tentatively assigned. In addition, the assignments of Rydberg states of SiH4 reported in the literature have been re-examined in light of the recently reported ionization potential [Berkowitz et al., J. Chem. Phys. 86, 1235 (1987)].

    Notes: High resolution photoabsorption cross section data for the CO 4PG (1,0) band have been obtained under several different temperature conditions. It was found that the absorption strength of the d 3Δ1–X 1Σ+ (5,0) band underlying the 4PG (1,0) band varies with temperature. The ratio of the absorption band strength of the d–X (5,0) band to that of the 4PG (1,0) band has been determined for temperatures at 150 K, 295 K, and 370 K. Adopting the integrated oscillator strength of the 4PG (1,0) from the recent literature we have determined band absorption cross sections for the two bands as a function of temperature. The ratios of the band absorption strengths of the A–X (1,0) and d–X (5,0) bands at different temperatures are compared with those calculated from the mixing coefficients determined by analysis of level shifts in high resolution spectra and lifetime measurements. © 1999 American Institute of Physics.

    Description: We have developed MGD007 (anti-glycoprotein A33 x anti-CD3), a DART protein designed to redirect T cells to target gpA33 expressing colon cancer. The gpA33 target was selected on the basis of an antibody-based screen to identify cancer antigens universally expressed in both primary and metastatic colorectal cancer specimens, including putative cancer stem cell populations. MGD007 displays the anticipated-bispecific binding properties and mediates potent lysis of gpA33-positive cancer cell lines, including models of colorectal cancer stem cells, through recruitment of T cells. Xenograft studies showed tumor growth inhibition at doses as low as 4 μg/kg. Both CD8 and CD4 T cells mediated lysis of gpA33-expressing tumor cells, with activity accompanied by increases in granzyme and perforin. Notably, suppressive T-cell populations could also be leveraged to mediate lysis of gpA33-expressing tumor cells. Concomitant with CTL activity, both T-cell activation and expansion are observed in a gpA33-dependent manner. No cytokine activation was observed with human PBMC alone, consistent with the absence of gpA33 expression on peripheral blood cell populations. Following prolonged exposure to MGD007 and gpA33 positive tumor cells, T cells express PD-1 and LAG-3 and acquire a memory phenotype but retain ability to support potent cell killing. In cynomolgus monkeys, 4 weekly doses of 100 μg/kg were well tolerated, with prolonged PK consistent with that of an Fc-containing molecule. Taken together, MGD007 displays potent activity against colorectal cancer cells consistent with a mechanism of action endowed in its design and support further investigation of MGD007 as a potential novel therapeutic treatment for colorectal cancer.
