    Notes: Abstract Purpose. GL331 is a new homolog of VP-16, and has demonstrated more efficacious anti-cancer activity in both the in vitro and in vivo lymphoma systems. To extensively explore GL331's clinical value, we furthermore evaluate the cytotoxicity and apoptosis-inducing activity of GL331 in several human cell lines from cancers that are not normally treated with VP-16. Methods. By MTT and clonogenic survival assays, the cytotoxicities of GL331 and VP-16 were evaluated in a variety of cell lines including nasopharyngeal, hepatocellular, gastric, colon, cervical, and neuro-blastoma cancer types. Western blot analysis was performed to detect the MDR-1 level in these cell lines. By Annexin V-staining flow cytometry and detection of DNA ladders, the apoptosis-inducing activities of GL331 and VP-16 were also evaluated. Results. GL331 showed more efficacy than its congener VP-16 in killing cancer cells. The estimated ID50 of GL331 were 2.5 to 17-fold lower than those of VP-16. GL331 possessed more cell-killing activity even in MDR-1-overexpressing cell lines such as HCC36 and SW620. Its higher cytotoxicity could be attributed by the elevated ability to induce apoptotic cell death. Conclusions. GL331's overriding drug resistance and higher cancer cell-killing activity suggest its superiority in clinical cancer therapy.

    Notes: Summary Clones from aFrankia At4 gene bank were pooled into groups and mass conjugated into anodD mutant ofRhizobium leguminosarum bv.viciae by triparental matings. When peas were inoculated with the pooled transconjugants, nodulation was observed. A plasmid, pAt2GX containingFrankia DNA, was isolated from bacteria recovered from these nodules. This plasmid was shown to complement anodD mutant ofR. leguminosarum bv.viciae. Thus pAt2GX contains aFrankia gene that is functionally equivalent tonodD ofR. leguminosarum bv.viciae.

    Notes: Actinoplanes missouriensis produces an intracellular soluble glucose Isomerase. The soluble enzyme can be purified by a DEAE-cellulose beads columm with a onestep salt elution. The purified enyzme exhibited a molecular weight of approximately 80,000 daltons, being composed of two identical subunits of about 42,000 daltons each. The Km for glucose is 1.33M, the Km for frucotse is 1.67M. The enzyme has an optimal pH of 7.0. The presence of the cobalt ion is not required to produce optimal activity of the enzyme if the proper amount of magnesium is present.

    Notes: Candida rugosa lipase has been used to investigate the hydrolysis of high concentration olive oil in the AOT-isooctane reversed micellar system at Wo = 10, pH 7.1, and 37°C. Results from this work show the hydrolytic reaction obeys Michaelis-Menten kinetics up to the initial substrate concentration of 1.37M, with turnover number kcat and Michaelis constant KM of 67.1 μmol/min mg enzyme and 0.717M, respectively. A competitive inhibition by the main product, oleic acid, has been found with a dissociation constant KI for the complex EP* of 0.089M. The rate equation was further analyzed in the time course reaction and was found in agreement with the experimental results for lower substrate concentrations, up to 0.341M. Large deviation occurred at high substrate concentrations, which may be due to the effects of large consumption of water on kinetics, on the formation of glycerol, and on the deactivation of lipase in the hydrolysis reaction as well.

    Notes: Terminating arterial vessels, the structure of sinuses and cords, and the passage of cells through the sinus wall in the red pulp of human spleen were studied. All terminating arterial capillaries arterial capillaries observed opened into cords. The distance between terminating arterial capillaries and sinuses varied. Macrophages were commonly present at arterial terminations. Arterial capillary endothelium contained filaments about 80 Å in diameter. Blood cells were frequently present in the capillary lumen or in passage through the capillary wall into cords. Endothelial cells of sinuses contained three distinctive structures: loosely organized cytoplasmic filaments, tightly organized finer filaments, and micropinocytotic vesicles. Many micropinocytotic vesicles about 0.1 μ in diameter were present just beneath the plasma membrane of the lateral and luminal sides of sinus endothelial cells and a few at the basal aspect. Loosely organized filaments about 80 Å in diameter ran parallel to the longitudinal axis of the sinus endothelium. The finer filaments about 30-50 Å in diameter were tightly organized as filamentous bands and present basally. The filaments of the bands appeared inserted upon the plasma membrane. They were also present in the cordal reticular cells and terminating arterial capillaries. Free cells were frequently present in passage through the slits of the sinus wall. There were no preformed or fixed apertures in the sinus wall. The basement membrane and reticular fibers were completely covered by the endothelial cells and/or cordal reticular cells.It is likely that those slits between endothelial cells in the sinus wall not covered by the basement membrane are potential passageways for cells moving from the cords into the sinus. The larger cytoplasmic filaments are likely contractile. The filamentous bands appear to maintain cell shape, stabilize the wall in relation to the basement membrane, and are probably operative in the control of cellular passage through the slits of sinus wall.

    Notes: The spleens of rats were studied by light and electron microscopy during the course of an acute myelogenous leukemia, with special reference to infiltration of leukemic myeloblasts in the spleens and to the correlation of leukemic cell infiltration with splenic hematopoiesis and splenomegaly. Leukemic myelo-blasts infiltrated the cordal space of the red pulp. Many of them appeared in groups. Even in spleens which were heavily infiltrated, leukemic myeloblasts did not penetrate the white pulp. Massive infiltration and proliferation of the leukemic myeloblasts in the red pulp resulted in splenomegaly. The spleen increased its he-matopoietic activity, while the medullary hematopoiesis diminished due to the invasion of leukemic myeloblasts in the bone marrow. Compensatory splenic hemato-poiesis occurred in most of the leukemic spleens, but diminished in spleens which were very heavily infiltrated with leukemic myeloblasts. Thus, the degree of splenomegaly and splenic hematopoiesis did not necessarily correspond to the percentage of leukemic myeloblasts in the bone marrow, but rather related to the number of leukemic myeloblasts present in the spleen. A possible role for the splenic sinus walls in promoting compensatory hematopoiesis in the spleen is discussed. A consistent association of type “C” virus particles with leukemic myeloblasts was observed.

    Notes: The porous crosslinked poly(vinyl alcohol) beads with the L-proline pendant was synthesized as the chiral stationary phase (CSP) for ligand-exchange chromatography of amino acid racemates via suspension polymerization of vinyl acetate and triallyl isocyanurate as a crosslinker, methanolysis of the copolymer, glycidylation of the formed crosslinked poly(vinyl alcohol), and final functionalization with L-proline. After the polymer with the chiral ligand was complexed with copper(II) cations, it was employed as the CSP to resolve the enantiomers of 15 amino acids. The results showed that the CSP possessed powerful enantioselectivity and all the tried amino acid racemates were completely separated. The large loadability (up to 60 mg per injection) permitted its application on a large scale. © 1996 John Wiley & Sons, Inc.

    Notes: In this research, the RDX yield from nitrolysing hexamethylenetetramine (hexamine, HA) together with methylenedinitramine (MDNA) in 90% nitric acid was more than 100%. It was found that MDNA decomposed completely in 90% nitric acid, forming O(CH2ONO2)2, while in HNO3—NH4NO3 system the decomposed fragments of MDNA condensed into RDX. On the basis of the comparison of the experimental results to the 1H-NMR spectra obtained by following the decomposition reaction of MDNA with nitric acid, the intermediate, HOCH2NHNO2, was postulated as one of the possible precursors to RDX both in Hale and in K process.
