    Notes: Abstract Shuttle mutagenesis using signature-tagged transposons was employed to generate a library of individually tagged mutants of the Neisseria meningitidis strain B1940, which belongs to serogroup B. The use of tagged transposons allowed us to monitor for enrichment for single mutants during the process of shuttle mutagenesis, by amplification of the tags and subsequent sequence determination. Enrichment of a single clone occurred during the transformation of the meningococci with transposon-containing plasmid DNA. Sequence determination around the site of transposon insertion revealed that the transposon had mutagenized a previously unknown locus, which was designated hrtA (high rate of transformation). hrtA-mediated transformation was independent of TnMax5 and tag sequences, and it most probably involved recombination events. The hrtA locus is restricted to meningococci and gonococci and is present in few apathogenic neisserial species. Chromosomal mapping of hrtA and six further hrt sites revealed a random distribution of highly transforming DNA fragments on the meningococcal chromosome. In conclusion, our data demonstrate that shuttle mutagenesis of naturally competent bacteria using signature-tagged transposons allows the isolation of chromosomal DNA fragments, which exhibit a high transformation efficiency, and which, therefore, are likely to be involved in horizontal gene transfer.

    Description / Table of Contents: Summary To assess the epidemiological situation of salmonellosis in Germany a total of 1608 strains of salmonella sampled in a sentinel-study were isolated and typed. The results on occurrence, distribution, types and antbiotic resistance of salmonella were compared with both the strains forwarded to the National Reference Center (NRC) for Salmonellosis for typing from all regions of Germany and the strains notified according to the duty of notification in the five new Länder (federal countries) and Berlin. In all three studies about 40% of salmonellosis were observed between July and September. Approximately a quarter of salmonellosis fell into the age group 0 to 5 years, around half of salmonellosis occurred in the age groups 0 to 14 years. The incidence rates in the sentinel-study and of the notified salmonellosis were highly corresponding with rates of 160.8 and 165.4, respectively. S. enteritidis and S. typhimurium were the most important serovars. Among the S. enteritidis strains the rates of resistance to the antimicrobial agents used were just 0–2.6%. Also the rate of multiresistant strains of S. enteritidis was very low, 1.3% and 2.8%, respectively. By contrast among the S. typhimurium strains high rates of resistance to ampicillin (A), chloramphenicol (C), streptomycin (S), sulfonamide (Su), tetracycline (T) were found. The drug resistance of S. typhimurium DT 104 had a special significance (R-type ACSSuT). About 90% of the strains tested showed this pattern of drug resistance. Among the other salmonella only the resistance to tetracycline (20%) and to sulfonamide (80%) played a role. The fact is important that up to now among all salmonella resistance to ciprofloxacin (0.2%) and cefotaxim (0.1%) was observed rarely. The dominant epidemic strain S. enteritidis PT 4 was not declining and it was not replaced by the emerging epidemic strains S. typhimurium DT 194 so far.
    Notes: Zusammenfassung Zur Einschätzung der Salmonellose in Deutschland 1997 wurden im Rahmen der Sentinel-Studie 1608 Salmonella-Stämme isoliert und typisiert. Die erzielten Ergebnisse über Vorkommen, Verbreitung, Typenspektrum und Antibiotikaresistenz der Salmonellen wurden mit den dem Nationalen Referenzzentrum aus allen Regionen Deutschlands vorliegenden humanen Salmonella-Isolaten sowie stellvertretend für das gesamte Bundesgebiet mit den 29 326 nach BSeuchG aus den neuen Bundesländern und Berlin gemeldeten Salmonellen verglichen. In allen drei Studien wurden zwischen Juli und September etwa 40% der Salmonellosen beobachtet. Entsprechend lagen in diesem Zeitraum die monatlichen Inzidenzraten zwischen etwa 20 und 30%. Etwa ein Viertel der Salmonellosen entfiel auf das Säuglings-, Kleinkind- und Vorschulalter, die knappe Hälfte auf die Altersklasse bis 14 Jahre. S. enteritidis und S. typhimurium waren die mit Abstand wichtigsten Serovare. Bei S. enteritidis war das Resistenzniveau gegen fast alle getesteten Antibiotika mit Resistenzanteilen zwischen 0 bis 2,6% sehr niedrig. Multiresistente S. enteritidis-Stämme waren mit Anteilen von 1,3% bzw. 2,8% zu vernachlässigen. Im Gegensatz dazu zeigte S. typhimurium eine beachtliche Resistenz gegen Ampicillin (A), Chloramphenicol (C), Streptomycin (S), Sulfonamide (Su), Tetracycline (T), besondere Bedeutung hat dabei vor allem die Antibiotikaresistenz von S. typhimurium DT 104. Wichtig ist, daß bisher bei Salmonellen nur vereinzelt Resistenzen gegen Ciprofloxacin (0,2%) und Cefotaxim (0,1%) beobachtet wurden. Prognostische Einschätzungen der vergangenen Jahre, daß der führende Epidemiestamm S. enteritidis PT 4 rückläufig ist und durch einen neuen aufkommenden Epidemiestamm S. typhimurium DT 104 ersetzt wird, haben sich 1997 nicht erfüllt.

    Description / Table of Contents: Zusammenfassung Zur numerischen Integration steifer retardierter Differentialgleichungen wurde ein stetiges einfach-implizites Runge-Kutta-Verfahren implementiert. Dieses implizite Runge-Kutta-Verfahren wurde auf Grund von Stabilitätsuntersuchungen einer großen Klasse von Interpolations-Integrations-Verfahren ausgewählt. Die numerischen Ergebnisse zeigen die Effektivität dieser Verfahren sowohl an Beispielen steifer gewöhnlicher wie retardierter Differentialgleichungen.
    Notes: Abstract A continuous singly-implicit Runge-Kutta method is implemented for stiff retarded differential equations. The choice of this implicit Runge-Kutta method is based on stability investigations of wide classes of interpolationintegration schemes. The numerical results show the effectiveness of these methods for both stiff ordinary and retarded differential equations.

    Description / Table of Contents: Zusammenfassung Bei der Radiotherapie periorbital gelegener Tumoren ist eine Strahlenschädigung des Auges oft unvermeidlich. In 26 Fällen wurden die Reaktionen am Auge nach Oberflächendosen, die zwischen 4320 und 14320 R betrugen, verfolgt und mit den histologischen Ergebnissen von 8 enucleierten Bulbi zusammengestellt. Abgesehen von der Sklera wiesen alle Augengewebe Veränderungen auf. Im Vordergrund standen schwere Hornhautprozesse, auffallend häufig war das Strahlenglaukom. Die therapeutischen Möglichkeiten bleiben symptomatisch und wenig aussichtsreich. Nach Belastungen über 8000 R ist mit dem Verlust des Bulbus zu rechnen.
    Notes: Summary Damage to the eye is frequently unavoidable following radiotherapy of periorbital tumors. The reactions of the eye to radiation have been studied in 26 patients following surface-doses varying between 4320 and 14320 R. The results have been compared with the histological findings in 8 cases in which the eye was enucleated. Alterations were observed in all tissues excepting the sclera. Serious corneal changes were the most impressing observation. The radiation glaucoma was very frequent. Therapeutic possibilities are limited to symptomatic treatment and little promising. A loss of the bulb is to be expected following exposure to more than 8000 R.

    Notes: Abstract A cDNA clone of a defence response transcript was isolated from a library prepared from barley leaves expressing papilla resistance towards the powdery mildew fungus, Blumeria (syn. Erysiphe) graminis f.sp. hordei (Bgh). The 904 bp sequence encodes a 229 amino acid polypeptide with a putative signal peptide of 23 amino acids. After cleavage, the protein has a mass of 22.3 kDa and exhibits up to 60% amino acid identity to certain dicot proteins, and 46% amino acid identity to barley oxalate oxidase; therefore we designated it HvOxOLP (for Hordeum vulgare oxalate oxidase-like protein). Single-base substitutions among several cDNA and RACE clones demonstrate a gene of many copies. Both the transcript and protein accumulate from 3 h after inoculation with Bgh. The transcript level peaks at 18–24 h and subsequently decreases, whereas the protein level is stable from 24 h after inoculation. The accumulation patterns are independent of the outcome of the barley/powdery mildew interaction, unlike that of PR proteins, for example. The transcript accumulates specifically in the inoculated epidermal tissue. This temporal and spatial expression pattern suggests a very close relationship to papilla formation. Immunoblot analyses have facilitated a demonstration that HvOxOLP, like oxalate oxidase, is a water-soluble 100 kDa oligomeric protein. The oligomer is heat-stable and SDS-tolerant, and it can be denatured into a 25 kDa monomer. Attempts to demonstrate oxalate oxidase activity for this protein have failed. However, the relationships to oxalate oxidase suggests that HvOxOLP may be involved in H2O2 generation necessary for, for example, cross-linking of cell wall components during formation of papillae.

    Notes: Abstract Stable transformants of the human hepatoma cell line HepG2 were established that constitutively transcribe a DNA unit consisting of a stretch of hepatitis B virus DNA and of nonviral DNA conferring resistance to neomycin. Previously it had been shown that upon cotransfection of such transformants with replication-competent HBV DNA, transcripts of such units become reverse transcribed, demonstrating that DNA constructs can function as defective replication units. Transformed cell lines stably transcribing the defective replication units could be shown to use the transcriptional starts for the viral pregenome and for the large core antigen at a ratio of 9:1. Upon the induction of replicative processes in the transformed cells by transfection with replication-competent wild type (wt) DNA, defective pregenomes transcribed from the integrated state became included in the pool of replicating nucleic acids.
