    Notes: Summary yc4 is an unstable yellow-in-the-dark mutation found in the unicellular green alga Chlamydomonas reinhardtii. yc4 reverts to the wild-type green-in-the-dark phenotype at the frequency of 1x10−4 revertants per cell division. We have tested whether mutation and reversion of yc4 is due to the insertion and excision of the C. rein-hardtii transposon TOC1. Southern blots showed differences in the distribution of TOC1 elements when the original mutant yc4 strain, which contained over 30 TOC1 elements, was compared with green-in-the-dark-revertants. However, analysis of the yc4 mutation in a different genetic background containing half the number of TOC1 elements showed that reversion of yc4 is not associated with loss of TOC1 elements.

    Notes: Abstract Sixty-four cases of theophylline poisoning were reviewed. All but two cases represented intentional self poisoning. The majority of patients were young females who presented acutely after ingestion of sustained release preparations prescribed for asthma. Serum theophylline levels (mean 365 μmol/l, SD 177) indicated a high risk of toxicity. Electrolyte and metabolic abnormalities (hypokalaemia, hypomagnesaemia, hypophosphataemia, hyperglycaemia, acid-base disturbances and leucocytosis) were common. Serum potassium, serum glucose, leucocyte count and length of stay in the intensive care unit all correlated strongly with maximum serum theophylline level (p〈0.001). The low incidence of life-threatening manifestations of severe toxicity (hypotension, serious arrhythmias or seizures) and excellent outcome, contrasts with many previous reports. The results support the use of a management regimen which emphasizes intensive supportive therapy and restricts the use of charcoal haemoperfusion.

    Description / Table of Contents: Zusammenfassung Aufnahme und Einbau von 14C-Ölsäure in Phospholipide, Triglyzeride und Cholesterin-Ester von Fettstreifen (fatty streaks) und Atheromen (fibrofatty lesion) in menschlichen Femoralarterien wurde untersucht und mit Aufnahme und Einbau in die genannten Lipidfraktionen in benachbarten normalen Intima-Bezirken und in die Media derselben Arterien verglichen. In Fettstreifen kommt es zu einem vermehrten Einbau von 14C-Ölsäure in Triglyzeride und Phospholipide im Vergleich zu der benachbarten normalen Intima und der darunter liegenden Media. Der Einbau in Cholesterin-Ester ist besonders hoch in Fettstreifen (ungefähr 20mal) verglichen mit der angrenzenden normalen Intima. Andererseits ist der vermehrte Einbau von Ölsäure in die drei Lipidfraktionen bei Atheromen nicht sehr ausgeprägt. Die Bedeutung des Lipidstoffwechsels in der Entstehung der frühen menschlichen atherosklerotischen Veränderungen wird diskutiert.
    Notes: Summary The uptake and incorporation of (1-14C)-oleic acid into phospholipid, triglyceride and cholesterol ester by fatty streak and fibro fatty lesions in human femoral arteries was investigated and compared with the uptake and incorporation into these lipid fractions in normal intima and media in the same arteries. In fatty streak lesions, increased incorporation of (1-14C)-oleic acid into triglyceride and phospholipid occured in relation to the adjacent normal intima and media. Incorporation into cholesterol ester is particularly high (approximately 20 times) in fatty streak lesions compared with the adjacent normal intima. On the other hand, increased incorporation of oleic acid into the three lipid fractions is not marked in the fibro fatty lesion. The significance of metabolism of lipid in the pathogenesis of the early human lesions is discussed.

    Notes: Summary Twenty-four hours after nerve crush, the Schwann cell plasma membrane and subjacent outer layer of Schwann cell cytoplasm were examined by freeze-fracture in myelinated fibres from the rat and rabbit sciatic nerves. The irregular circumferential bands and longitudinal columns of cytoplasm which characterise the surface of the normal Schwann cell were diminished or had disappeared. Concomitantly, the membrane pores (of micropinocytotic vesicles or caveolae) which are normally present on the bands and columns were also lost. The loss of these specialised features is discussed in terms of the onset of cell dedifferentiation in association with cell division. The Schwann cell plasma membrane acquired a new feature: deep impressions of adjacent collagen fibres, present particularly on the cytoplasmic bands and columns and presumed to be related to a transient increase in fibre diameter.

    Notes: Summary Using the freeze-freeze-fracture technique, the sciatic nerve of the rat and rabbit was examined distally at 24 h after crush, with particular reference to the node of Ranvier and paranode. The paranodes, in the majority of myelinated fibres, showed a loss of the cytoplasmic circumferential bands and longitudinal columns and their associated membrane pores which characterise the normal Schwann cell surface. Axonal changes consisting of accumulations of axoplasmic organelles occurred at both the node and paranode. At the nodes large intramembraneous partiles in the axolemma (E face) appeared unchanged. Nodal Schwann cell microvilli and paranodal myelin terminal loops were generally unaffected. The findings are discussed in terms of the decrease in amplitude of the action potential which occurs in early Wallerian degeneration.

    Notes: Background : Poor compliance with 5-aminosalicylic acid therapy has been reported amongst patients with inflammatory bowel disease. Currently, there is no easy method to monitor 5-aminosalicylic acid; however, the chemical similarity between 5-aminosalicylic acid and salicylate might provide a solution.Aim : To determine the feasibility of using salicylate levels to monitor compliance with 5-aminosalicylic acid medication.Methods : Thirty-six patients with inflammatory bowel disease, taking maintenance 5-aminosalicylic acid, provided either a paired serum and urine sample or an intestinal biopsy. Samples were split into two: half were sent to the hospital biochemistry department for salicylate measurement, and half were analysed for 5-aminosalicylic acid and its metabolite, N-acetyl-5-aminosalicylic acid, using high performance liquid chromatography. Correlation between the results was calculated.Results : Serum and urine were available for 25 patients. Serum salicylate was undetectable, but urinary salicylate ranged from 31 to 3254 µg/mL. The correlations between urinary salicylate and 5-aminosalicylic acid and N-acetyl-5-aminosalicylic acid were 0.96 (95% confidence interval, 0.91–0.98) and 0.9 (95% confidence interval, 0.77–0.96), respectively. Sixteen biopsies were available from 13 patients. The 5-aminosalicylic acid and N-acetyl-5-aminosalicylic acid concentrations were 0.2–657 ng/mg and 1.6–1598 ng/mg, respectively; there was no correlation with bowel salicylate.Conclusions : The close correlation between 5-aminosalicylic acid and salicylate levels offers a simple method to assess compliance with 5-aminosalicylic acid therapy.

    Notes: Abstract We attempted to determine whether Chlamydomonas reinhardtii telomeres, which do not form G-quartet structures readily in vitro, are able to nucleate telomere addition in Saccharomyces cerevisiae. Restricted C. reinhardtii genomic DNA was ligated to a linear S. cerevisiae vector lacking a telomere. A C. reinhardtii telomere ligated to this unprotected end allowed vector replication as a linear DNA molecule in S. cerevisiae. DNA sequencing revealed common [T4AG3]n and variant T6AG3 and T5AG3 C. reinhardtii telomere repeats capped by S. cerevisiae telomere repeat units. The recognition of a C. reinhardtii telomere by the telomere maintenance machinery of S. cerevisiae is consistent with a common theme for telomere structure in organisms with divergent telomere repeats.
