    Description / Table of Contents: Summary It is demonstrated by analysis of four standard reference materials (NBS 1632 a coal, NBS 1633 a coal fly ash, NBS 1648 urban particulate matter and BCR 176 city waste incineration ash) that the selenium determination by Solid Sampling ZAAS works well, if fine grained elemental nickel is used as matrix modifier. Optimum nickel-to-sample ratios are in the range of 2∶1 to 8∶1. Admixture of graphite powder is necessary in some cases in addition. The detection limit is about 0.1 μg/g. If the matrix is well-known, the described method is a convenient and fast alternative to conventional trace analytical techniques.
    Notes: Zusammenfassung Durch die Analyse von vier Standardreferenzmaterialien (NBS 1632 a Kohle, NBS 1633 a Kohleflugasche, NBS 1648 Stadtstaub und BCR 176 Stadtmüllverbrennungsasche) wird gezeigt, daß die Selenbestimmung mittels Feststoff-ZAAS zu guten Ergebnissen führt, wenn feinkörniges elementares Nickelpulver als Matrixmodifizierer eingesetzt wird. In der Regel ist ein zwei- bis achtfacher Nickelüberschuß erforderlich, ggf. auch eine Beimischung von Graphitpulver. Die Nachweisgrenze des Verfahrens liegt bei etwa 0,1 μg/g. Bei gut bekannter Matrix ist die beschriebene Methode eine bequeme und schnelle Alternative zu konventionellen spurenanalytischen Techniken.

    Notes: Summary Addition of fine grained elemental nickel stabilizes selenium in inorganic solid samples, so that no selenium losses can be detected up to 2,000 °C pretreatment temperature. The specific absorption signals of selenium are significantly broadened and delayed. Interferences by sulphur are overcome by this effect, because the sulphur compounds appear in the gas phase before selenium. For organic materials, however, the addition of nickel as matrix modifier for solid sampling ZAAS fails.

    Description / Table of Contents: Summary A report is given on the successful application of direct ZAAS as a trace-analytical tool for inorganic materials characteristically occurring in cement industries. Results of interlaboratory studies and analysis of standard reference materials for important elements as As, Cd, Cr, Hg, Tl and others are discussed in detail. Examples show that trace analysis directly from the solid is often successful using aqueous solutions for calibration. Characteristic interferences of direct ZAAS could normally be eliminated by admixture of graphite powder.
    Notes: Zusammenfassung Es wird über den erfolgreichen spurenanalytischen Einsatz der direkten ZAAS an für die Zementindustrie characteristischen anorganischen Proben berichtet. Dabei stehen die Ergebnisse von Ringversuchen und die Analyse von Standardreferenzmaterialien für so wichtige Elemente wie As, Cd, Cr, Hg, Tl und anderen im Mittelpunkt der Untersuchungen. Anhand von Beispielen wird gezeigt, daß die direkte Spurenbestimmung aus dem Festkörper häufig mit wäßrigen Bezugslösungen erfolgreich durchgeführt werden kann. Charakteristische Störungen der Direkten ZAAS konnten in der Regel durch Graphitbestimmungen beseitigt werden.

    Notes: Abstract  Background: Multidrug resistance (MDR) describes the phenomenon of cross-resistance between different cytostatic agents which are structurally and functionally dissimilar. Two recently discovered proteins, lung resistance protein (LRP) and the multidrug resistance-related protein (MRP) have been implicated in the development of MDR. Since resistance to chemotherapeutic agents is a common problem in filtration surgery, especially in cases of complicated glaucoma, we decided to investigate the presence of MRP and LRP in surgically removed Tenon specimens from glaucoma patients. Methods: The presence of MRP and LRP in surgically removed Tenon tissue (n=15) was analyzed by immunohistochemistry. The expression by cultured Tenon fibroblasts was assessed by reverse-transcriptase polymerase chain reaction (RT-PCR) and fluorocytometry. Results: LRP expression was detected in 8 of 10 Tenon specimens. Positive staining for MRP was obtained in 5 of 10 specimens. Negative controls with non-immune mouse IgG did not display any specific staining. RT-PCR and fluorocytometry revealed constitutive expression of MRP and LRP, at the RNA and protein level respectively, that was unaltered by pretreatment of the cells with mitomycin C or 5-fluorouracil. Conclusion: Our results demonstrate, that besides P-glycoprotein, other components of the MDR-system are present in conjunctival fibroblasts. Future developments in the use of chemotherapeutic agents in association with of filtration surgery need to take account of the presence of these counteracting mechanisms.

    Notes: Abstract Vitronectin, an integrinbinding a-l-glycoprotein with potent cell-adhesion and proliferation-mediating properties, has been shown to be incorporated in surgically removed membranes from patients with proliferative vitreoretinopathy (PVR), proliferative diabetic retinopathy (PDR) and macular pucker. Therefore we developed an ELISA technique to quantify levels of vitronectin in human vitreous and plasma samples in order to be able to evaluate the significance of vitronectin in these different vitreoretinal diseases. Our results indicate a significant increase of vitronectin in all proliferative disorders except idiopathic macular pucker. Adjustment of all probes to equal total protein content yielded a significant increase only in PDR (type 11 diabetes). Plasma samples demonstrated a significant increase of vitronectin in patients with type Il diabetes suffering from PDR. Therefore, break-down of the blood-retina barrier appears to be the most likely explanation for the increased levels of vitronectin in the vitreous. Our results indicate that vitronectin may not only be involved in the regulation of epiretinal membrane formation at significantly higher levels in patients with type II diabetes, but the increase of vitronectin in diabetic plasma may also help to explain the pathological alteration of the coagulation cascade in diabetics.
