    Abstract: Background Oligodeoxynucleotides (ODNs) containing unmethylated cytosine-guanosine (CpG) sequence motifs constitute the immunostimulatory components of bacterial DNA which potently activate innate immunity. Administration of CpG-ODNs before the onset of experimental colitis prevents intestinal inflammation by induction of colitis-suppressing T cells. Aims To identify the interplay between innate and adaptive immune cells finally leading to protective CpG-ODN effects in intestinal inflammation. Methods Total splenic cells or purified selected cell types (CD4(+)CD62L(+) T cells alone or with B cells or dendritic cells (DCs)) from BALB/c mice were (co)-incubated in vitro with CpG-ODN for 5 days and CD4(+)CD62L(+) cells were injected intraperitoneally into C.B.-17 SCID (severe combined immunodeficiency) mice. Splenic CD4(+)CD62L(+) T cells were isolated from transgenic donor mice in which CD11c(+) DCs were depleted by diphtheria toxin administration during CpG-ODN treatment and injected into C57BL/6 Rag2(-/-) recipients. Intestinal inflammation was evaluated by histological scoring and cytokine secretion of mesenteric lymph node cells. Results CpG-ODN treatment of total splenic cells but not of purified CD4(+)CD62L(+) cells reduced the colitogenic potential of transferred T cells. While CpG-ODN stimulation of co-cultured CD4(+)CD62L(+) and B-cells did not alter the colitogenic potential of T cells, co-incubation of CpG-ODN-stimulated DCs and CD4(+)CD62L(+) cells reduced the colitogenic potential of the T cell population. Depletion of CD11c(+) DCs during CpG-ODN administration in vivo abolished the protective CpG-ODN effects. Conclusions CpG-ODN-dependent protective effects in experimental colitis act indirectly on CD4(+)CD62L(+) T cells. While the involvement of B cells could be excluded, CD11c(+) DCs were identified as key mediators of CpG-ODN-induced protection in experimental colitis.

    Abstract: The clinical outcome of spinal cord injury (SCI) depends in part on the extent of secondary damage, to which apoptosis contributes. The CD95 and tumor necrosis factor (TNF) ligand/receptor systems play an essential role in various apoptotic mechanisms. To determine the involvement of these ligands in SCI-induced damage, we neutralized the activity of CD95 ligand (CD95L) and/or TNF in spinal cord-injured mice. Therapeutic neutralization of CD95L, but not of TNF, significantly decreased apoptotic cell death after SCI. Mice treated with CD95L-specific antibodies were capable of initiating active hind-limb movements several weeks after injury. The improvement in locomotor performance was mirrored by an increase in regenerating fibers and upregulation of growth-associated protein-43 (GAP-43). Thus, neutralization of CD95L promoted axonal regeneration and functional improvement in injured adult animals. This therapeutic strategy may constitute a potent future treatment for human spinal injury

    Notes: Zusammenfassung In den Jahren 1952–1956 wurden 78 Kinder wegen Morbus hämolyticus neonatorum oder dessen Folgen stationär in die Grazer Universitäts-Kinderklinik aufgenommen, davon 46 Kinder mit Unverträglichkeit im Rh-System oder deren Untergruppen und 31 von Müttern mit Isoimmunisierung im Bereich der klassischen Blutgruppen. 16 der Rh-Fälle und 7 der AB0-Kinder wiesen eine Mitbeteiligung des ZNS im Sinne eines Kernikterus auf. Weiters Mitteilung der Krankengeschichten von 5 frühgeborenen Kindern und 2 ausgetragenen Neugeborenen mit klinisch bzw. pathologisch-anatomisch nachweisbarem Kernikterus ohne Anhalt für irgendeine Immunisierung. Auf Grund der Erfahrungen der letzten Jahre muß festgestellt werden, daß der Kernikterus ganz allgemein im Zunehmen begriffen ist, wobei einerseits die Immunisierung im Bereiche der klassischen Blutgruppen an Bedeutung gewinnt, andererseits aber auch solche Fälle von Kernikterus sich mehren, die ätiologisch vorerst nicht zu klären sind. Beim Zustandekommen der nervösen Hirnveränderungen scheint es sich um einen komplexen Mechanismus zu handeln, wobei die durch Funktionsausfall der Leber hervorgerufene Hyperbilirubinämie wohl die wichtigste Rolle spielen dürfte. Diese Annahme bestimmt auch unser therapeutisches Vorgehen dahingenhend, daß vorerst bei allen Neugeborenen mit Icterus gravis auf der Grundlage eines Morbus hämolyt. neonat. bei Überschreiten eines Blutspiegels an indirektem Bilirubin von 15 mg-% die Durchführung eines Blutaustausches geboten ist. Hingegen scheint die Wirkung eines solchen bei Frühgeborenen mit Kernikterus eher ungünstig zu sein. An Hand des eigenen Krankengutes werden aus dem bunten Krankheitsbild der Kernikterusfolgen auch weniger bekannte Symptome, wie: Krämpfe, Muskelhypotonie mit Muskelatrophie und schließlich Veränderungen an den Augen in Form der Opticusatrophie herausgegriffen und etwas eingehender besprochen. Abschließend wird auf die Tatsache hingewiesen, daß zahlreiche Kernikterusfolgen im Laufe der Zeit einer spontanen Besserung fähig sind.

    Notes: Objectives: Bacterial cultivation is a well-established method for analyzing plaque samples. Real-time polymerase chain reaction (PCR) is a novel rapid method for the identification and quantification of periodontopathogenic bacteria that has been recently introduced. In this study, we compared real-time PCR with conventional anaerobic cultivation.Method: A total of 78 subgingival plaque samples were harvested from pockets 〈inlineGraphic alt="geqslant R: gt-or-equal, slanted" extraInfo="nonStandardEntity" href="urn:x-wiley:03036979:JCPE740:ges" location="ges.gif"/〉5 mm in 22 patients with advanced chronic periodontitis and immediately transferred into transport medium. Aliquots were evaluated with species-specific probes by real-time PCR (meridol® Perio Diagnostics, GABA) and anaerobic bacteria culture on selective media for the detection of Actinobacillus actinomycetemcomitans, Fusobacterium nucleatum, Porphyromonas gingivalis, Prevotella intermedia and Tannerella forsythensis. The analysis was performed by two separate, blinded examiners.Results: When real-time PCR was compared with the culture method (golden standard) for the detection of putative periodontopathogenic bacteria, the sensitivity and specificity for A. actinomycetemcomitans were 67% and 100%, respectively (κ: 0.79); for F. nucleatum 73% and 53%, respectively (κ: 0.21); for P. gingivalis 94% and 84%, respectively (κ: 0.77); for P. intermedia 33% and 94%, respectively (κ: 0.26) and for T. forsythensis 92% and 56%, respectively (κ: 0.51). Spearman's correlation coefficients for the quantitative results of both methods were 0.82 for A. actinomycetemcomitans, 0.33 for F. nucleatum, 0.83 for P. gingivalis, 0.38 for P. intermedia and 0.67 for T. forsythensis.Conclusion: Overall, the agreement between both test methods was excellent for A. actinomycetemcomitans and P. gingivalis, fair for T. forsythensis and poor for F. nucleatum and P. intermedia. The discrepancies in the results may be explained by the inability of cultivation methods to distinguish between close related taxa, and the problems of keeping periopathogenic bacteria viable, which is required for standard cultivation.
