    Description / Table of Contents: Résumé Les auteurs, dans le cadre d'une étude ultrastructurale du foie foetal humain, ont utilisé une méthode directe à pH 8 pour mettre en évidence l'activité de la phosphatase alcaline. L'accumulation du produit final de la réaction histochimique (phosphate de plomb) a pu être observée de façon constante sur les membranes plasmatiques des hepatocytes et des érythroblastes. Dans la lignée érythropoïétique les dépôts sont toujours plus intenses sur les cellules les plus immatures. Les auteurs ont observé également des dépôts importants de phosphate de plomb dans certains ≪granules≫ péribiliaires ainsi que dans des ≪granules≫ des cellules érythropoïétiques. La disposition des dépôts le long des membranes cellulaires et dans certains organites cytoplasmiques est discutée en fonction de la technique de mise en évidence.
    Notes: Summary A direct lead method for visualization of alkaline phosphatase activity at pH 8 was used in an electron microscope study of the foetal development of human liver. An accumulation of lead phosphate, the final product of the enzymatic reaction, was observed on the cell membranes of hepatocytes and also on those of erythroblasts, especially in immature cells. An intense reaction was observed in peribiliary granules and in granules of the erythropoietic cells. The localization of lead phosphate precipitate along the cell membranes and in some cytoplasmic bodies is discussed in relation to the procedure used.

    Notes: Summary A systematic study of an autofluorescence method is described to improve the early histological diagnosis of myocardial ischaemia. Our results on 732 autopsy cases including 182 cases of sudden death show that the autofluorescence examination of haematoxylin and eosin stained sections of the myocardium is not only reliable in the identification of recent ischaemic lesions, but contributes to a better histological evaluation. In 24 cases undetected by white light examination it allowed recognition of ischaemic lesions.

    Notes: Summary In one case of Dubin-Johnson Syndrom, the Authors call attention to the importance of mitochondrial alterations, which involve more than half of these organelles. These alterations are of 2 different types: the first one is a fibrillary degeneration, as previously published in many other cases, and the second a vacuolary degeneration of a new type. The hepatocyte also shows an alteration of its sinusoidal surface and an accumulation of biliary pigments.

    Notes: The phenotype of 165 gastrointestinal stromal tumours was studied by immunohistochemistry. In each case the phenotype was compared to the histological diagnosis. The phenotype was muscle in 49 tumours (30%), neural in 18 (11%), histiocytic in 20 (12%) and mixed in five (3%); 68 tumours (41%) were positive for vimentin only, four tumours had no markers and one tumour was positive for keratin only. Histologically, the tumours were classified as smooth muscle, probably smooth muscle, probably nerve sheath tumours or tumours of undetermined differentiation, In 30 istologically unequivocal muscle tumours, the phenotype was muscle in 28. Half of them, all benign, arose in the oesophagus or gastric cardia. Apart from this group, there was no correlation between phenotype, site of tumour and histological differentiation. Actin was a more sensitive muscle marker than desmin. With the exception of oesophageal tumours, the histological appearances alone could not establish a diagnosis of malignancy and were inadequate in evaluating differentiation. Immunohistochemical examination determined differentiation in 54% of the tumours, but this finding should be interpreted with caution in terms of histogenesis. It allowed us. however, to specify the differential diagnosis in 57 tumours in which the histological diagnosis was uncertain.

    Notes: Forty-three cases of large cell anaplastic thyroid carcinoma were examined with various antisera. Four histological patterns were identified: spindle cell, giant cell, trabecular and squamous. In 38 cases the epithelial origin was demonstrated with various epithelial markers: 11 cases stained positively for thyroglobulin, 19 for T3 and/or T4, 35 for fat globule membrane antigens, 28 for keratin, 29 for lactoferrin, and one for calcitonin. Five cases were negative for all epithelial markers but could not be characterized further since, except for vimentin, they did not have mesenchymal markers. The immunohistochemical proof of the epithelial nature of anaplastic thyroid tumours is given by staining with anti-keratin and anti-fat globule membrane antigen. In addition the detection of lactoferrin seems useful, but not that of thyroglobulin which was present in only 4% of the spindle cell tumours and in 32–56% of the other histological types. Thirty carcinomas were positive for vimentin; the co-expression of vimentin and epithelial markers seems frequent in thyroid anaplastic carcinomas.

    Description / Table of Contents: Résumé Les auteurs, étudiant les diverses cellules épithéliales rencontrées dans la région périportale du foie foetal humain, décrivent deux types d'éléments qui possèdent des caractères ultrastructuraux intermédiaires entre ceux de la cellule épithéliale biliaire et ceux de l'hépatocyte. Ils décrivent un type cellulaire à cytoplasme et noyau fortement denses, différents de ceux que l'on observe dans les «dark cells» de Steiner et coll. (1962b).
    Notes: Summary The fine structure of the different cells at the margin of the portal spaces was investigated in the foetal human liver. Two types of cells intermediate between biliary epithelial and parenchymal elements were described. In addition, a type of cell in which both the cytoplasm and nucleus are electron-dense was observed. This element is different from the dark cell of Steiner et al. (1962b).
