    Abstract: Centrioles are essential for ciliogenesis. However, mutations in centriole biogenesis genes have been reported in primary microcephaly and Seckel syndrome, disorders without the hallmark clinical features of ciliopathies. Here we identify mutations in the genes encoding PLK4 kinase, a master regulator of centriole duplication, and its substrate TUBGCP6 in individuals with microcephalic primordial dwarfism and additional congenital anomalies, including retinopathy, thereby extending the human phenotypic spectrum associated with centriole dysfunction. Furthermore, we establish that different levels of impaired PLK4 activity result in growth and cilia phenotypes, providing a mechanism by which microcephaly disorders can occur with or without ciliopathic features.

    Notes: Summary The properties of three different recombinant hepatitis B virus core proteins expressed inEscherichia coli were compared: an N-terminal fusion protein, a C-terminally truncated protein and a sequence-authentic protein. All three proteins assembled into capsid-like particles with typical HBc-antigenicity, sedimentation behavior and distinctive electron microscopical images. Apart from this, however, variant HBc proteins displayed properties different from sequence-authentic HBc protein p21.4. Unlike p21.4, the particles of the N-terminal fusion protein p22.2 were sensitive to proteolytic attack by trypsin at variable sites within its arginine-rich C-terminus but not in its extended N-terminus. We therefore conclude that the C-terminal region is located on the surface of the p22.2 particle. These particles also showed increased HBe-antigenicity, as did the C-terminally truncated core particles p17.6, and to an even greater extent p18* particles which were derived from p22.2 by tryptic digestion. This might be interpreted as evidence for an — albeit minor — structural change. All variant core particles were less stable and contained less RNA. Electron microscopic indication for DNA binding of C-terminal deleted p17.6 particles was obtained using an aqueous spreading technique.

    Description / Table of Contents: Summary A report on the further development of an automatic data acquisition and remote transmitting system is given. The improvements distinguishing the new device in comparision with the earlier one are described. Then a discussion of the errors and influencing effects follows. By means of a theoretical estimation it is tried to give a prediction on the system reliability which is to be expected. Finally the operation and the maintenance of the device as well as the possibilities in removing errors are treated shortly.
    Abstract: Résumé On rapporte ici le développement apporté à une installation automatique de mesures et de transmission. On décrit en particulier les améliorations qui distinguent l'appareillage actuel d'un ancien. On y ajoute une discussion des fautes de mesure et des divers éléments agissant sur le tout. Au moyen d'une estimation théorique, on essaie de prévoir la sécurité que l'on est en droit d'attendre du système. On décrit enfin brièvement le service et l'entretien de l'installation ainsi que les possibilités d'éliminer les erreurs.
    Notes: Zusammenfassung Es wird über die Weiterentwicklung einer automatischen Meßwerterfassungs- und-übertragungsanlage berichtet. Die Verbesserungen, in denen sich die jetzige Anlage gegenüber der früheren unterscheidet, werden beschrieben. Dem schließt sich eine Diskussion der Meßfehler und Einflußgrößen an. Mittels einer theoretischen Abschätzung wird der Versuch unternommen, Voraussagen über die zu erwartende Systemzuverlässigkeit zu geben. Schließlich werden die Bedienung und die Wartung der Anlage sowie die Möglichkeiten der Fehlerbeseitigung kurz behandelt.

    Notes: The structure of jack bean chitinase was solved at 1.8 Å resolution by molecular replacement. It is an α-helical protein with three disulfide bridges. The active site is related in structure to animal and viral lysozymes. However, unlike in lysozyme, the architecture of the active site suggests a single-step cleavage. According to this mechanism, Glu68 is the proton donor and Glu90 assists in the reaction by moving towards the substrate and recruiting a water molecule that acts as the nucleophile. In this model, a water molecule was found in contact with Glu90 Oε1 and Thr119 Oγ at a distance of 3.0 and 2.8 Å, respectively. The model is in accordance with the observed inversion mechanism.
