    Description: Background/Aim: Lung cancer is the leading cause of cancer-related death and a better marker for advanced personalized therapeutic approaches, such as immunotherapies, is in urgent need. Interleukin-12 (IL-12) is a cytokine that has been reported to exhibit potent tumoricidal effects, however, the contribution of IL-12 genotypes to lung cancer is still largely unrevealed. The aim of this study was to investigate whether single nucleotide polymorphisms (SNPs) in IL-12A and IL-12B are associated with lung cancer in a Taiwanese population. Materials and Methods: Genotypes of 358 lung cancer patients and 716 controls were determined by the polymerase chain reaction–restriction fragment length polymorphism method. Results: The distributions of genotypic (p=0.0036) and allelic (p=0.0005) frequencies of IL-12A rs568408 demonstrated significant differences between cases and controls. In detail, the AA genotype of IL-12A rs568408 was associated with a significantly elevated risk of lung cancer compared with the GG genotype (odds ratio(OR)=2.41, 95% confidence interval(CI)=1.36-4.29, p=0.0021). No difference was observed regarding IL-12A rs2243115 and IL-12B rs3212227 genotypes between the case and control groups. In addition, the results of interaction analysis showed that the AA genotype of IL-12A rs568408 was associated with elevated lung cancer risk, especially among those with smoking habits (p=0.0043). Conclusion: IL-12A rs568404 AA genotype may contribute to the etiology and serve as a genomic determinant of lung cancer in Taiwanese, especially smokers.

    Description: Self-renewal is the hallmark feature both of normal stem cells and cancer stem cells. Since the regenerative capacity of normal haematopoietic stem cells is limited by the accumulation of reactive oxygen species and DNA double-strand breaks, we speculated that DNA damage might also constrain leukaemic self-renewal and malignant haematopoiesis. Here we show that the histone methyl-transferase MLL4, a suppressor of B-cell lymphoma, is required for stem-cell activity and an aggressive form of acute myeloid leukaemia harbouring the MLL-AF9 oncogene. Deletion of MLL4 enhances myelopoiesis and myeloid differentiation of leukaemic blasts, which protects mice from death related to acute myeloid leukaemia. MLL4 exerts its function by regulating transcriptional programs associated with the antioxidant response. Addition of reactive oxygen species scavengers or ectopic expression of FOXO3 protects MLL4(-/-) MLL-AF9 cells from DNA damage and inhibits myeloid maturation. Similar to MLL4 deficiency, loss of ATM or BRCA1 sensitizes transformed cells to differentiation, suggesting that myeloid differentiation is promoted by loss of genome integrity. Indeed, we show that restriction-enzyme-induced double-strand breaks are sufficient to induce differentiation of MLL-AF9 blasts, which requires cyclin-dependent kinase inhibitor p21(Cip1) (Cdkn1a) activity.     Description: Polarization is one of the basic properties of electromagnetic waves conveying valuable information in signal transmission and sensitive measurements. Conventional methods for advanced polarization control impose demanding requirements on material properties and attain only limited performance. We demonstrated ultrathin, broadband, and highly efficient metamaterial-based terahertz polarization converters that are capable of rotating a linear polarization state into its orthogonal one. On the basis of these results, we created metamaterial structures capable of realizing near-perfect anomalous refraction.     Description: Background: Despite age and sex differences in fecal hemoglobin (f-Hb) concentrations, most fecal immunochemical test (FIT) screening programs use population-average cut-points for test positivity. The impact of age/sex-specific threshold on FIT accuracy and colonoscopy demand for colorectal cancer screening are unknown. Methods: Using data from 723,113 participants enrolled in a Taiwanese population-based colorectal cancer screening with single FIT between 2004 and 2009, sensitivity and specificity were estimated for various f-Hb thresholds for test positivity. This included estimates based on a "universal" threshold, receiver-operating-characteristic curve–derived threshold, targeted sensitivity, targeted false-positive rate, and a colonoscopy-capacity-adjusted method integrating colonoscopy workload with and without age/sex adjustments. Results: Optimal age/sex-specific thresholds were found to be equal to or lower than the universal 20 μg Hb/g threshold. For older males, a higher threshold (24 μg Hb/g) was identified using a 5% false-positive rate. Importantly, a nonlinear relationship was observed between sensitivity and colonoscopy workload with workload rising disproportionately to sensitivity at 16 μg Hb/g. At this "colonoscopy-capacity-adjusted" threshold, the test positivity (colonoscopy workload) was 4.67% and sensitivity was 79.5%, compared with a lower 4.0% workload and a lower 78.7% sensitivity using 20 μg Hb/g. When constrained on capacity, age/sex-adjusted estimates were generally lower. However, optimizing age/-sex-adjusted thresholds increased colonoscopy demand across models by 17% or greater compared with a universal threshold. Conclusions: Age/sex-specific thresholds improve FIT accuracy with modest increases in colonoscopy demand. Impact: Colonoscopy-capacity-adjusted and age/sex-specific f-Hb thresholds may be useful in optimizing individual screening programs based on detection accuracy, population characteristics, and clinical capacity.     Description: High-density polyethylene (HDPE)/ultra-high-molecular-weight polyethylene (UHMWPE) fibre composites were prepared via solution crystallization to investigate the components of epitaxial crystal growth on a highly oriented substrate. Scanning electron microscopy morphologies of HDPE crystals on UHMWPE fibres revealed that the edge-on ribbon pattern crystals that were formed initially on UHMWPE fibres converted afterwards to a sheet shape as crystallization progressed. Wide-angle X-ray diffraction confirmed that the polymer chain oriented along the fibre axis and the orthorhombic crystal form of HDPE remained unchanged in HDPE/UHMWPE fibre composite systems. The thermal behaviour of the fibre composites measured by differential scanning calorimetry showed double melting peaks, the nature of which, as disclosed by partial melting experiments, is ascribed to bilayer components existing in the induced crystals: the inner layer is composed of more regularly folded chain crystals induced by UHMWPE fibres, and the outer layer formed on the inner one with a thinner and lower ordered crystal structure.

    Description: Rpc34 is a subunit of the Rpc82/34/31 subcomplex residing on the DNA-binding cleft of RNA polymerase (Pol) III. Rpc34 contains a structurally flexible N-terminal tandem winged-helix (tWH) domain related to the TFIIE transcription factor. While the second WH (WH2) fold of the tWH domain is known to function in DNA melting activity during transcription initiation, the functional role of the WH1 fold is unknown. In this study, we generated a series of new Rpc34 tWH mutants conferring a cold-sensitive growth phenotype. We found that the tWH mutations severely compromised in vitro transcription activity due to destabilization of the preinitiation complex (PIC). Site-specific protein photo-cross-linking analysis indicated that the tWH domain persistently interacts with protein subunits of the Pol III cleft in the PIC and the ternary elongation complex (TEC). Furthermore, purified Pol III proteins with tWH mutations also showed reduced efficiency in RNA elongation. Our study results suggest that the tWH domain is an important protein module above the Pol III cleft that integrates protein and nucleic acid interactions for initiation and elongation.

    Notes: Initiation of chemical reactions by ultra-violet light is characterized by essentially complete lack of temperature dependence and by physical separation of the source or initiating agent from the reaction medium. The advantage of these features for reactor stability and control are discussed in a theoretical study of the photo-sensitized polymerization of styrene in a continuous stirred tank reactor and a comparison is made with the behavior of the thermally initiated reaction.

    Notes: Summary A theoretical analysis is presented to investigate the thermal interaction between laminar film condensation of a saturated vapor and a forced convection system separated by a heat conducting wall. In this work, the effect of the wall thermal resistance is considered. It is assumed that the countercurrent boundary layer flow is formed on the two sides. Governing boundary layer equations together with their corresponding boundary conditions for film condensation and forced convection are all cast into dimensionless forms by using the non-similarity transformation. The resulting system of equations is solved by using the local non-similarity method in conjunction with the fourth order Runge-Kutta method in conjunction with the Nachtsheim-Swigert iteration scheme. The total heat flux through the wall and the wall temperature distribution are determined. The present results show that the effect of the forced convection Prandtl number Pr c is not negligible for large values of the thermal resistance ratioA *, and the effect ofA * and Pr c on the overall heat transfer through the wall is more pronounced than that of the Jakob number and film Prandtl number.

    Notes: Good-quality high-Tc YBa2Cu3O7-x thin films deposited by the laser ablation method were investigated by Raman spectrosocpy. The vibrational spectra of thin films of YBa2Cu3O7-x at various laser intensities and with different polarizations were measured. The Raman modes change on increasing the laser power and in general such changes are reversible. However, as the laser intensity is increased above a certain value, dramatic changes occur in the Raman peaks, as a result of damage to the sample.
