Notes: Abstract Fumarase-free electron particles from Propionibacterium freudenreichii and P. pentosaceum were prepared by discontinuous sucrose gradient centrifugation, and the influence of 2-n-heptyl-4-hydroxy-quinoline-N-oxide (HQNO) and ultraviolet irradiation on the reduction of menaquinone and cytochrome b with l-lactate or glycerol-3-phosphate and the reoxidation by fumarate was studied. In the presence of HQNO the steady state reduction level of menaquinone during fumarate reduction was increased whereas the steady state reduction level of cytochrome b was decreased as compared with the reduction levels measured in the absence of HQNO. The steady state reduction level of menaquinone during electron transport to fumarate was not influenced by ultraviolet irradiation and the steady state reduction level of cytochrome b was decreased at increasing irradiation times. The data indicate that cytochrome b is involved in the electron transport to fumarate.

Notes: During incubation of potato tuber discs (Solanum tuberosum L. cv. Bintje) on a callus-inducing medium at 28°C, a high capacity of in vivo alternative pathway respiration develops (75% of uninhibited respiration is azide-resistant). When callus induction takes place at 8°C, only 45% of respiration is resistant to azide. In the lag phase of growth the activity of alternative pathway is low. during the exponential growth phase the activity reaches its maximal rate. This in vivo activity is of the same size at both culture temperatures. As a consequence a greater part of alternative pathway capacity is operating in uninhibited respiration during growth at low temperatures.

Notes: ATP content (per g fresh weight) and energy charge were higher during incubation at 8°C than at 28°C in the early stages of callus induction of potato tuber discs (Solanum luberosum L. cv. Bintje). After a transfer from 28°C to 8°C, ATP content increased while a sharp decline in ATP content was observed after a transfer from 8°C to 28°C. ADP and AMP pools did not increase correspondingly. When the callus discs had entered the logarithmic growth phase, the energy charge was maintained within relatively narrow limits (0.77–0.80) at all culture temperatures.

Notes: Summary LDH is localized along various intracytoplasmatic membranes of osteoclasts. Macrophages show membrane-bound LDH only after phagocytosis of calcium hydroxylapatite, the main mineral constituent of bone. The localization of LDH in these macrophages is almost the same as in osteoclasts. The significance of this finding, and its possible implication in the process of bone resorption and the origin of osteoclasts are discussed.
