    Description: The epidermal growth factor receptor (EGFR) is a member of the erbB family of receptors and is overexpressed in many tumor types. A repebody is a newly designed nonantibody protein scaffold for tumor targeting that contains leucine-rich repeat modules. In this study, 3 64 Cu-labeled anti-EGFR repebodies with different chelators were synthesized, and their biologic characteristics were assessed in cultured cells and tumor-bearing mice. Methods: Repebodies were synthesized with the chelators 2-( p -isothiocyanatobenzyl)-1,4,7-triazacyclononane- N,N',N,''- triacetic acid trihydrochloride ([ p -SCN-Bn]-NOTA), 2,2',2''-(10-(2-(2,5-dioxopyrrolidin-1-yloxy)-2-oxoethyl)-1,4,7,10-tetraazacyclododecane-1,4,7-triyl) triacetic acid (DOTA- N -hydroxysuccinimide ester), or 1-( p -isothiocyanatobenzyl)diethylenetriamine pentaacetic acid trihydrochloride ([ p -SCN-Bn]-DTPA) in 1.0 M NaHCO 3 buffer (pH 9.2) for 24 h. Purified NOTA-, DOTA-, and DTPA-conjugated repebody were radiolabeled with 64 Cu in 0.1 M NH 4 OAc buffer (pH 5.5). To compare the EGFR-binding affinities of the repebodies, cellular uptake studies were performed with the human non–small cell lung cancer cell line H1650 (high expression of EGFR) and the human colon adenocarcinoma cell line SW620 (low expression of EGFR). Biodistribution and small-animal PET imaging studies were performed using H1650 tumor–bearing mice. Results: Radiochemical yields of the 64 Cu-labeled repebodies were approximately 70%–80%. Cellular uptake of the NOTA-, DOTA-, and DTPA-repebodies was over 4-fold higher in H1650 cells than in SW620 cells at 1 h. The 3 repebodies had accumulated specifically in H1650 tumor–bearing nude mice by 1 h after intravenous injection and were retained for over 24 h, as measured by the percentage injected dose per gram of tissue (%ID/g). Tumor uptake of all repebodies increased from 1 to 6 h (at 1 h, 6.28, 8.46, and 6.91 %ID/g for NOTA-, DOTA-, and DTPA-repebody, respectively; at 6 h, 9.4, 8.28, and 10.1 %ID/g, respectively). H1650 tumors were clearly visible after injection of each repebody, with high tumor-to-background ratios (at 1 h, 3.43, 4.89, and 2.38 for NOTA-, DOTA-, and DTPA-repebody, respectively; at 6 h, 3.05, 4.36, and 2.08; at 24 h, 3.81, 4.58, and 2.86). Conclusion: The 3 64 Cu-repebody complexes demonstrated specific and rapid uptake in EGFR-expressing tumors within 1 h and may have potential as novel EGFR imaging agents for PET.

    Notes: Abstract A 2.3 kb DNA fragment that contains a gene encoding endoinulinase, inu2, from Aspergillus ficuum ATCC 16882 was isolated and analyzed. It includes an open reading frame of 1,551 bp, coding for a polypeptide with calculated molecular weight of 55,790 Da, including a putative signal peptide of 22 amino acids. Alignment of amino acid sequences revealed 73.3% identity and 93.9% similarity between A. ficuum and Penicillium purpurogenum endoinulinase.

    Notes: Abstract Restriction pattern, chromosome localization and the sequence of the testis-specific gene,TSPY, were investigated in the white-handed gibbon, agile gibbon, siamang, hamadryas baboon and Japanese monkey. Southern blot analysis showed theTSPY gene to be male specific in the primates used and disclosed variability of restriction pattern in gibbons. Fluorescencein situ hybridization demonstrated that the probe ppTSPY2372, biotinylated using polymerase chain reaction, is located as a slight signal in the proximal long arm of the Y chromosome of the white-handed gibbon, hamadryas baboon and Japanese monkey and in the middle long arm of the Y chromosome of the siamang, while a faint signal and an intense signal were detected in the proximal long arm of the Y chromosome of the agile gibbon. These findings allow us to speculate that the gibbons might have evolved some structural differentiation in theTSPY gene. The first introns of theTSPY genes were sequenced and compared. One hundred thirty-seven of 606 sites were found to be variable, and 10 deletions/insertions were noted among these gibbons, two species of Old World monkeys and human. Sequence similarity ranged from 81.7% between humans and hamadryas baboons to 98.7% between Japanese monkeys and hamadryas baboons. These sequences may be of great use in future studies for resolving the phylogeny of gibbons and Old World monkeys.

    Notes: Abstract Yarrowia lipolytica KCCM50506, which transforms isobutyric acid to L-β-hydroxy isobutyric acid (L-β-HIBA), was screened. Chemostat cultures were carried out in jar fermentors at dilution rates of 0.02 h−1 to 0.12 h−1. L-β-HIBA fermentation-regulating factors were determined to be specific growth rate, and concentrations of glucose and isobutyric acid in fermentor from analysis of steady-state data. The specific productivity of L-β-HIBA increased as the specific growth rate increased, apparently as a growth-associated type of product formation. A fed-batch culture was carried out under optimum conditions where the concentrations of glucose and isobutyric acid in the fermentor were maintained at 23 g l−1 and 9 g l−1, respectively. The concentrations of cells and L-β-HIBA obtained at the end of fermentation were 20 g l−1 and 49 g l−1, respectively, corresponding to 2.0 and 2.7 times more than concentrations in batch culture.

    Notes: Summary The purpose of this paper is to examine subjects to show the nature and limit of interdisciplinary communication in existing environmental programmes in US graduate schools. Ultimately, this analysis may provide more effective communication with the general public. Following comparative historical reviews of both the sciences and the human activity for environmental protection, and a content analysis of empirical documents used by the public, a mushroom computer model has been produced. This model is based on organizational behaviour theory and contains the following 15 subjects which were originally introduced before the lack of communication resulting from ‘curriculum failure’ in the 19th century: philosophy, politics, economics, architecture, sociology, biology, medicine, agriculture, ecology, public health, mathematics, physics, chemistry, geology, statistics. The subjects should foster effective communication with the general public, leading to more effective environmental protection.

    Notes: Abstract With the application of early reperfusion therapy after acute MI, the incidence and importance of nontransmural infarction is increasing. In a rat model with nontransmural infarction, we evaluated 1) the changes of LV dimension, LV interstitial fibrosis and transforming growth factor-β1 (TGF-β1) mRNA expression and 2) the effects of angiotensin converting enzyme (ACE) inhibitor and angiotensin II type 1 (AT1) receptor antagonist treatment. Female Sprague-Dawley rats were subjected to 45 min of coronary occlusion followed by reperfusion, and five days after the operation the animals were randomized to untreated (n = 19), captopril-treated (n = 15) and losartan-treated (n = 14) groups. Twenty-six days after MI, echocardiographic examination revealed a remarkable dilatation of LV. Captopril or losartan treatment reduced the extent of LV cavity dilatation. Collagen volume fractions in noninfarcted septum as well as in peri-infarct area decreased with captopril or losartan treatment, compared to those of the untreated rats. In noninfarcted septum of untreated rats, TGF-β1 mRNA expression increased more than two fold (P 〈 0.05 vs. pre-MI) 5 and 10 days after MI. Captopril or losartan treatment suppressed the acute induction of TGF-β1 mRNA expressions. These results indicate that ACE inhibitor or AT1 receptor antagonist treatment after nontransmural infarction 1) attenuates LV remodeling as in transmural infarction and 2) decreases interstitial fibrosis at least partly by blocking the acute induction of TGF-β1 mRNA expression.

    Notes: Summary A white rot fungus was isolated from rotted leaves and identified as Pestalotiopsis sp. KCTC 8637P. It produced a flocculant biopolymer. A flocculant was partially purified from the culture broth by series of precipitations with 95% ethanol and named as Pestan. The components of Pestan were consisted of glucose : glucosamine : glucuronic acid : rhamnose with a approximately molar ratio of 100:3.5:1.6:1.3. In kaolin suspension(final concentration was 4,800 mg/l), the highest flocculating activity was attained at the biopolymer flocculant concentration of 1 mg/l . The flocculating activity was observed most highest by the addition of cationic solutions, especially 8mM CaCl2 · 2H2O or 8mM FeCl3. The thermal stability of Pestan was sustained up to 70 °C.
