    Abstract: Urothelial umbrella cells are characterized by apical, rigid membrane plaques, which contain four major uroplakin proteins (UP la, Ib, II and III) forming UPla/UPII and UPIb/UPIII pairs. These integral membrane proteins are thought to play an important role in maintaining the physical integrity and the permeability barrier function of the urothelium. We asked whether the four uroplakins always coexpress in the entire human lower urinary tract. We stained immunohistochemically (ABC-peroxidase method) paraffin sections of normal human ureter (n = 18) and urinary bladder (n = 10) using rabbit antibodies against UPIa, UPIb,. UPII and UPIII; a recently raised mouse monoclonal antibody (MAb), AU1 and two new MAbs, AU2 and AU3, all against UPHIII; and mouse MAbs against umbrella cell-associated cytokeratins CK18 and CK20. Immunoblotting showed that AU1, AU2 and AU3 antibodies all recognized the N-terminal extracellular domain of bovine UPIII. By immunochemistry, we found that in 15 cases of human ureter, but in only 2 18 TO cases of bladder, groups of normal-looking, CK18-positive umbrella cells lacked both UPIII and UPIb immunostaining. The UPIb/UPIII-negative cells showed either normal or reduced amounts of UPla and UPII staining. These data were confirmed by double immunofluorescence microscopy. The distribution of the UPlb/UPIII-negative umbrella cells was not correlated with localized urothelial proliferation (Ki-67 staining) or with the distribution pattern of CK20. Similar heterogeneities were observed in bovine but not in mouse ureter. We provide the first evidence that urothelial umbrella cells are heterogeneous as some normal-looking umbrella cells can possess only one, instead of two, uroplakin pairs. This heterogeneity seems more prominent in the urothelium Of human ureter than that of bladder. This finding may indicate that ureter urothelium is intrinsically different from bladder urothelium. Alternatively, a single lineage of urothelium may exhibit different phenotypes resulting from extrinsic modulations due to distinct mesenchymal influence and different degrees of pressure and stretch in bladder versus ureter. Additional studies are needed to distinguish these two possibilities and to elucidate the physiological and pathological significance of the observed urothelial and uroplakin heterogeneity.     Description / Table of Contents: Zusammenfassung Aus Crotonöl konnten durch Anwendung multiplikativer Verteilungs- und chromatographischer Verfahren die entzündlichen und cocarcinogenen Substanzen A1–A4 und B1–B7 rein dargestellt werden. Diese Substanzen sind Diester des polyfunktionellen Diterpenalkohols Phorbol, C20H28O6 mit jeweils einer kurzkettigen (Essig-, (+)-S-2-Methylbutter-, Tiglinsäure) und einer langkettigen Fettsäure (Capryl-, Caprin-, Laurin-, Myristin-, Palmitinsäure). Die Teilsynthese einiger Substanzen wird angegeben. Aus den UV-, IR- und KMR-Daten und chemischen Befunden werden Teilformeln für Phorbol abgeleitet und diskutiert.
    Notes: Summary The compounds A1–A4 and B1–B 7 have been isolated from croton oil in pure state by multiplicative distribution and chromatography. These compounds are di-esters of the polyfunctional diterpene alcohol phorbol C20H28O6 each with a short chain (acetic-, (+)-S-2-methyl-butanoic-, tiglic acid) and a long chain fatty acid (octanoic-, decanoic-, dodecanoic-, tetradecanoic-, hexadecanoic acid). The partial synthesis of some compounds is described. From ultraviolet-, infrared-, nmr-spectra and from chemical evidence partial structures for phorbol are being discussed.

    Description / Table of Contents: Zusammenfassung Der Stoffwechsel der radioaktiv markierten und tumorpromovierenden Phorbolester TPA und PDD, sowie des zu PDD epimeren und inaktiven 4α-PDD, wurde 12 Std nach topischer Applikation auf die Rückenhaut von Mäusen untersucht. Die Ergebnisse werden mit dem Stoffwechsel von Phorbolderivaten nach in vitro Inkubation mit embryonaler Mäusehaut und mit Zellkulturen von HeLa-sowie L-Zellen verglichen. Wie bereits früher im Falle von TPA festgestellt, werden in der Mäusehaut auch PDD und 4α-PDD innerhalb von 12 Std kaum metabolisch verändert. In Monolayer-und Suspensions-Kulturen von HeLa-Zellen, wo nur die tumor-promovierenden Phorbolderivate bereits in sehr niedrigen Konzentrationen (10−8M) die DNA-Synthese blockieren und den Einbau von Cholin in Phospholipide stimulieren, wurden nur geringe metabolische Veränderungen der getesteten Phorbolderivate beobachtet. Nach Inkubation mit Monolayer-und Suspensionskulturen von l-Zellen, die diese Beeinflussung des DNA-und Lipidstoffwechsels durch Phorbolderivate nicht zeigen, werden dagegen alle hier untersuchten Phorbolderivate innerhalb von 12 Std in erheblichem Ausmaß metabolisiert. Die erhobenen Befunde sprechen gegen eine metabolische Aktivierung von Phorbolestern.
    Notes: Summary The metabolism of the radioactively labeled tumor promoting phorbol esters (TPA and PDD) and of the inactive epimer of PDD (4α-PDD) were studied 12 h after topical application on the back skin of mice. The results obtained were compared with metabolic changes of the labeled phorbol derivatives after in vitro incubation with embryonic mouse skin and with cultures of HeLa and L-cells. In mouse skin PDD and 4αPDD are poorly metabolized within 12 h as was shown previously for TPA. In monolayer and suspension cultures of HeLa cells where at very low concentrations (10−8M) only phorbol esters with tumor promoting activity in mouse skin block DNA synthesis and stimulate choline incorporation into lipids, hardly any metabolic changes were observed in biologically active as well as in inactive phorbol derivatives. In monolayer and suspension cultures of L-cells all phorbol derivatives tested are metabolized to a considerable extent within 12 h, which correlates with the lack of response in blocking DNA synthesis and stimulation of lipid turnover. From these results it is concluded that tumor promoting phorbol esters need no metabolic activation.

    Description / Table of Contents: Summary Intraperitoneal injections of polyinosinic acid/polycytidylic acid (poly I/C) reduce the tumor yields in Berenblum-experiments, if the synthetic, double-stranded ribonucleic acid is administered during tumor initiation of mouse skin by intragastric application of 7,12-dimethylbenz[a]anthracene. A reduction of tumor yield is not observed, if similar poly I/C treatment is scheduled before or during tumor promotion induced by topically administered phorbol ester TPA.
    Notes: Zusammenfassung Intraperitoneale Injektionen von Polyinosinsäure/Polycytidylsäure (Poly I/C) vermindern die Tumorausbeuten im Berenblum-Experiment, wenn die synthetische doppelsträngige Ribonucleinsäure während der Initiierungsphase der Mäusehaut—induziert durch intragastral verfüttertes 7,12-Dimethylbenz[a]anthracen — appliziert wird. Eine Verminderung der Tumorausbeuten durch ähnliche Poly I/C-Behandlung vor oder während der Promotionsphase — induziert auch topisch applizierten Phorbolester TPA — wird nicht beobachtet.

    Description / Table of Contents: Summary Procedures are described for specific tritum labeling of some highly active tumor promoting phorbol esters and for corresponding biologically inactive analogues. TPA-acetyl-3H (croton oil factor A1) with 6.3–100 mC/mmole is obtained following acetylation of 12-O-tetradecanoyl-phorbol-20-acetate and 12-O-tetradecanoyl-phorbol-20-tritylether in 13-position with acetic-anhydride-3H and removal of the protecting groups in 20-position. TPA-20-3H (croton oil factor A1), the tumor promotor PDD-20-3H and the corresponding biologically inactive 4α-epimer 4α-PDD-20-3H as well as phorbol-20-3H are prepared by seletive reduction of appropriate 20-aldehydes with sodium borohydride-3H. The specific activities of the labeled compounds range between 1.35 and 7.5 C/mmole. As intermediates of the reaction sequences developed for preparation of the labeled compounds some new derivatives of phorbol are described.
    Notes: Zusammenfassung Es werden Verfahren zur spezifischen Tritiummarkierung von tumorpromovierenden Phorbolestern mit hoher biologischer Aktivität sowie der entsprechenden inaktiven Derivate beschrieben. TPA-acetyl-3H (Wirkstoff A1, aus Crotonöl) wird mit einer spezifischen Aktivität von 6,3–100 mc/mmol erhalten, wenn man in 12-O-Tetradecanoylphorbol-20-acetat bzw. in 12-O-Tetradecanoyl-phorbol-20-trityläther die freie Hydroxylgruppe 13 mit Acetathydrid-3H verestert und danach die Schutzgruppe an OH-20 entfernt. TPA-20-3H (Wirkstoff A1 aus Crotonöl) der partialsynthetische Tumorpromotor PDD-20-3H und das entsprechende inaktive Epimere 4-α-PDD-20-3H sowie Phorbol-20-3H werden durch selektive Reduktion der entsprechenden 20-Aldehyde mit Natriumborhydrid-3H synthetisiert. Die spezifischen Aktivitäten der markierten Verbindungen liegen zwischen 1,35 und 7,5 C/mmol. Bei der Markierung als Zwischenprodukte anfallende neue Porbolderivate werden beschrieben.

    Notes: A continuous flow system with an enzyme electrode and with a new type of measuring instrument, the BIOXY-Meter, is described as an example for the determination of glucose. The BIOXY-Meter combined with various biosensors may be used in biotechnology and in other fields for the solution of a number of analytical problems. Measuring principle is the reduction of oxygen. The peak of first derivation of the current-time-curve is proportional to oxygen consumption and to decrease of substrate concentration. Enzyme fixation with common adhesive and a device for automated saturation of measuring samples with air in the sampler are also described. With the apparatus 60 samples per hour are analyzed for glucose. The linear measuring range is 0.1 mMol to 1.5 mMol with a coefficient of variation of Vk = 0.93 per cent. The described type of enzyme electrode is stable for 2 to 12 weeks or 1,000 to 5,000 measurements.
