    Description: The genetics of renal cancer is dominated by inactivation of the VHL tumour suppressor gene in clear cell carcinoma (ccRCC), the commonest histological subtype. A recent large-scale screen of approximately 3,500 genes by PCR-based exon re-sequencing identified several new cancer genes in ccRCC including UTX (also known as KDM6A), JARID1C (also known as KDM5C) and SETD2 (ref. 2). These genes encode enzymes that demethylate (UTX, JARID1C) or methylate (SETD2) key lysine residues of histone H3. Modification of the methylation state of these lysine residues of histone H3 regulates chromatin structure and is implicated in transcriptional control. However, together these mutations are present in fewer than 15% of ccRCC, suggesting the existence of additional, currently unidentified cancer genes. Here, we have sequenced the protein coding exome in a series of primary ccRCC and report the identification of the SWI/SNF chromatin remodelling complex gene PBRM1 (ref. 4) as a second major ccRCC cancer gene, with truncating mutations in 41% (92/227) of cases.     Notes: Abstract An electrochemical reactor packed with stainless steel fibres was used to oxidize a complexed copper cyanide solution. The reactor reduced the cyanide content from over 2000 p.p.m. to less than 100 p.p.m. in 4h. The energy consumption (kWh (kg CN−)−1) for a bipolar setup was larger than for the parallel setup. An analog circuit analysis qualitatively agrees with the experimental results.

    Notes: Abstract Adding a magnetic field gradient to the conventional TG system constructs the magnetic thermogravimetry analysis (TG(M) i.e. Faraday methods) and the magnetic derivative thermogravimetry (DTG(M)) techniques. We used the techniques to study the nanocrystalline processes of the FeCuNbSiB and FeCuNbCoSiB amorphous alloys. Some problems of their applications such as the characteristic temperature Tmin and TC are also discussed in detail.

    Notes: Abstract The σ—T and dσ/dT—T curves of the FeCuNbSiB amorphous alloy, which are the relationship between the total saturated magnetic moment per unit mass σ and temperature, are investigated by magnetic thermogravimetry analysis (TG(M)) technique. It is found that the crystallization process of the samples can be divided into five stages. The studies of samples annealed in temperature range of 480–610°C for 1h show that when the annealing temperature (Ta) is less than 540°C, the quantity of nanocrystalline α-Fe(Si) phase increases evidently with Ta, and the Curie temperature (TC) of residual amorphous phase also increases linearly with Ta, i.e. TC=0.52Ta+91.7°C, with correlation coefficient γ=0.98. The variation of volume fraction of α-Fe(Si) nanocrystalline phase or residual amorphous phase with Ta is measured by TG(M) technique.

    Notes: Abstract Poly (methacrylic acid) gels (PMAA gels) of various degrees of crosslinking were prepared and the dissociation behavior of these gels was examined; the swelling behavior was investigated as a function of the solution pH values. A reentrant phenomenon of swelling was observed and interpreted based on the Flory-Huggins equation and the Donnan equilibrium formula. Moreover, adsorption of L-lysine, oligo(L-lysine)s (Lys-n,n=3, 9, and 19) and poly(L-lysine) onto PMAA gels from aqueous solutions was investigated under different conditions of pH and concentration of adsorbate. The adsorption ratio of L-lysine onto PMAA gel is dependent on both the pH of solution and the degree of crosslinking. In a pH range between 8 and 9, the protonated form of L-lysine is strongly adsorbed on the PMAA gel by electrostatic interactions. Oligomers and polymer of L-lysine are adsorbed in a somewhat different way from the monomeric L-lysine. In addition, the desorption behavior of L-lysine from PMAA gels by a change in pH was also investigated.

    Description: Background/Aim: Anti-cancer activity of 3,5,7-trihydroxyflavone (galangin) has been documented in a variety of cancer types; however, its effect on human nasopharyngeal carcinoma (NPC) cells remains undetermined. Materials and Methods: Human NPC cell lines were treated with galangin. Apoptosis was analyzed by assessing nuclear condensation, cleavage of pro-caspase-3 and poly ADP-ribose polymerase (PARP), and DNA fragmentation. Short hairpin RNA-mediated silencing of p53 was used for characterizing the role of p53 in the anti-cancer activity of galangin. Phosphatidylinositol 3-kinase (PI3K) inhibitor, protein kinase B (AKT) inhibitor, and ectopic expression of wild type p85α or p85α mutant lacking p110α-binding ability were utilized to confirm the involvement of PI3K/AKT inactivation in galangin-induced apoptosis. Results: Galangin induces apoptosis and S-phase arrest by attenuating the PI3K/AKT signaling pathway. Silencing of p53 did not block the anti-cancer activity of galangin on NPC cells. Conclusion: Galangin effects on apoptosis and S-phase arrest in NPC cells are mediated via interfering with the PI3K-AKT signaling pathway in a p53-independent manner.
