    Notes: Abstract Numerous alterations in hormonal secretion occur with aging. In general, these tend towards a disintegration of the normal cyclic secretory patterns resulting in lower total circulating levels. In addition, declines in receptors and postreceptor function further decreases the ability of the hormonal orchestra to maintain coordinated function throughout the organism. Clues to some of these age-related changes in humans may come from the study of simpler organisms where regulatory systems are known to modulate the aging process. In particular, the interactions among the environment, hormones, and insulin receptor genes have led to new insights into the genetic control of longevity and the development of syndrome X.

    Notes: Summary The purity of the investigational antibacterial Abbott-086719.1 (I) which is 8[(3S)-3-amino-1-pyrrolidinyl]-1-cyclopropyl-7-fluoro-9-methyl-4-oxo-4H-quinolizine-3-carboxylic acid hydrochloride is determined using gradient elution HPLC. The chromatographic separation was optimized and the chromatographic parameters critical to separation are discussed in detail. Most acceptable separations were achieved using an Alltima C18 column (5 μm) measuring 4.6 mm I.D.×15 cm with 0.01M–0.03M citrate eluents which were modified with acetonitrile or with mixtures of acetonitrile and methanol. Related substances in I were determinable to 0.05%. Repeatability (RSD values) for determining related substances at levels of 0.50 to 0.06% ranged from ±1.9 to ±10%. Determinations of I in 5% dextrose in water and in hydroxypropyl methylcellulose were made using the same column and a simple isocratic system. The determination of I was stability indicating with precision (RSD values) of ±0.5% to ±2.0% and good agreement with theory for formulations containing I at 1.0 to 95 mg/mL concentrations. Recoveries of I from the vehicles were quantitative and linearity of the detector response of I was demonstrated to at least 0.10 mg/mL.

    Notes: Abstract The validation of multiplex solid-phase fluorescent minisequencing of mitochondrial DNA (mtDNA) for use in forensic casework is presented. Validation included testing of the reliability and species specificity of the technique, analysis of mixed body fluid samples, analysis of samples and substrate controls from previous cases and somatic stability of mtDNA. Animal, bacterial and fungal species extracts were examined and the test did not show cross-reactivity with other species. Hair, blood, saliva, faeces and semen or vaginal samples were tested from five male and five female individuals. For all the samples tested, heteroplasmy was observed only at position 302/309.1. Body fluid mixtures (blood:saliva, semen:saliva, faeces:semen, vaginal:semen) and DNA:DNA mixtures were examined. In total, 189 mixtures were analysed of which one resulted in a hybrid profile consisting of peaks from each of the two donors. The semen fraction of the semen:saliva and vaginal:semen mixtures appeared to be concentrated in the supernatant fraction of the extract thus highlighting the need to extract both the pellet and supernatant fractions of a stain. Control samples, crime stains and their substrate controls from previous cases were examined. Of the 12 loci typed by minisequencing, 11 could be verified by comparison to results from the sequencing method currently in use for casework and no discrepancies were observed between the two. MtDNA minisequencing was found to be a reliable and reproducible technique and its rapid and discriminating nature make it particularly suitable as a screening technique.

    Notes: The Ziegler polymerization of butene-1 has been studied in detail, by means of the pressure bottle technique. Eight principal conditions of polymerization have been varied. The amount and properties of the polymer have been studied, with particular emphasis on the atactic polymer. The polymers are generally from 50 to 70% atactic. Yields of over 300 g. polymer per gram of triisobutyl aluminum and intrinsic viscosities of over 2.5 have been obtained. The rate of polymerization goes through a sharp maximum at a catalyst ratio (Al/Ti) of 1.5-1.7. The kinetics are first-order in monomer, with a sharp drop in the rate constant after 1-5 hr. of polymerization.

    Notes: The version of time-dependent perturbation theory commonly used in the calculation of the hyperpolarizabilities of organic molecules through an expansion over electronic excited states is briefly rederived by a direct method, simplified for application to the nonresonant case. The application of the method within the framework of semiempirical quantum theory and singly and doubly excited configuration interaction is described. A particular parametrization of the CNDO/S method (CNDO/VS) that has been successful in estimating molecular β values for polar aromatic molecules is reviewed. Applications of the method to the interpretation of the molecular and crystal second-order properties of 2-(α-methylbenzylamino)-5-nitropyridine (MBANP) and to the calculation of the γ tensor for various types of conjugated compounds are given.

    Notes: Summary A simple and quantitative method for the preparation and high performance liquid chromatography (HPLC) of N-acylated fluoroquinolones has been developed. The acylation procedure was performed with four different acid anhydrides and found to be applicable to several fluoroquinolones. The acylated derivatives were chromatographed directly on a Nucleosil C18 column without the necessity of further sample manipulation. Detector response for the N-acetyl derivatives of norfloxacin, ciprofloxacin, sarafloxacin, and temafloxacin was linear to at least 50 μg/mL. The retention behavior of the N-acyl derivatives can be predicted by the length of the carbon chain of the amide.

    Notes: Abstract  We studied the development of spatial contrast-sensitivity and temporal-frequency selectivity for neurons in the monkey lateral geniculate nucleus. During postnatal week 1, the spatial properties of P-cells and M-cells are hardly distinguishable, with low contrast-sensitivity, sluggish responses, and poor spatial resolution. The acuity of P-cells improves progressively until at least 8 months, but there is no obvious increase in their maximum contrast-sensitivity with age. The contrast sensitivity of M-cells is already clearly higher than that of P-cells by 2 months, and at 8 months of age this characteristic difference between M- and P-cells approaches the adult pattern. There is a major increase in responsiveness during the first 2 postnatal months, especially for M-cells, the peak firing rate of which rises fivefold, on average, between birth and 2 months. Many P-cells in the neonatal and 2-month-old animals did not give statistically reliable responses to achromatic gratings, even at the highest contrasts: this unresponsiveness of P-cells might result from low gain and/or chromatic opponency. The upper limit of temporal resolution in the neonate is low – about one-third of that in the adult. Among M-cells, the improvement in temporal resolution, like that in contrast sensitivity, is rapid over the first 2 months, followed by a slower change approaching the adult value by 8 months of age. The development of contrast sensitivity, responsiveness and temporal tuning are little affected, if at all, by binocular deprivation of pattern vision from birth for even a prolonged period.

    Notes: Summary Vibratory detection thresholds were measured at a number of frequencies between 5 and 320 Hz following a lesion of the lateral digital nerve innervating the terminal phalanx of the left index finger. Thresholds measurements began approximately 4 weeks after the nerve was repaired. A staircase method was used to determine thresholds on both the injured fingerpad and the intact fingerpad of the opposite hand. There was a large increase in thresholds on the injured fingerpad in the lower range of frequencies (5–40 Hz) while at higher frequencies (80–250 Hz) there was no significant difference between the thresholds on the injured fingerpad and those on the intact fingerpad. It is suggested that the differential effect of the nerve lesion on vibratory thresholds reflects the spread of the vibratory stimulus through the skin and the spatial characteristics of functionally intact receptor/afferent groups innervating neighbouring skin.
