    Notes: Abstract Current pricing and charging methods for the Internet are not based on actual usage of this service, which leads to unfairness and more important, it does not deliver the right signals through financial incentives to network providers to upgrade critical links of their networks. The development of new multimedia applications and the convergence to an integrated services network will foster the tremendous growth of the Internet even more. With the Next Generation Internet not only technical services like bandwidth reservation will be introduced, but also new applications will emerge within the Internet. Charging the Internet in a fashion that provides feedback to users and providers has been proposed since the early '90s, however, only a few implementations and real-world examples are known today. This is due to subsidizing the Internet in its early stages and due to a technical development that did not care much about charging. With the recent redesign of the Internet protocol suite and discussions on multiple service classes in the Internet, architectures for charging and accounting have to be revisited, too. Economic models for the Internet cannot be tested fully and validated in non-real-world environments, because of the unknown user behavior. With this uncertainty over what models and pricing schemes to choose, it is evident that a specific charging and accounting platform will never be accepted by the community. In this paper a novel and flexible architecture for charging and accounting is proposed that provides a wide range of mechanisms and lets researchers experiment in an environment as close as possible to the targeted system. As a first step, four different pricing schemes are described, qualitatively assessed on the proposed platform, and a prototypical implementation performed. One of the economic models that have been implemented on Arrow is based on different service classes including reservation and recalculates prices dynamically depending on the traffic situation.

    Notes: Summary We evaluate a method for improved, ice-crystal-free cryofixation of various biological materials (macromolecules, subcellular fractions, whole cells in suspensions or as monolayers) without any chemical pretreatments. The principle of the method (as introduced byPscheid, Schudt, andPlattner 1981 for cell monolayer cultures) is as follows. Firstly, a thin sample layer is sandwiched between a thin copper object holder and a thermally insulating plastic sheet (Thermanox, which is also used as a cell culture substratum); secondly, a jet of melting propane is shot from a simple, inexpensive brass vessel onto the copper object holder. We now ascertained that this simple and inexpensive method allows cooling rates up to 〉 18,000 °C/sec and, thus, gives better results than comparable but more sophisticated and expensive procedures. The method also avoids all the disadvantages of some other rapid freezing methods. Specifically, the sample mass required is minimal, the sample remains fully hydrated and no changes of ionic or other conditions can occur. There is no hazard of passing a cold gas phase (cooling curves being very steep right at the beginning) or of impairment by mechanical impact (as it could be with “cold metal block freezing”). The expedient of using a thermal insulator as part of a sandwich sample allows cooling rates which are superior or at least equivalent to those obtained with a commercial double propane-jet device (and double copper sandwiches). The reason for this is that in practice a two-side propane jet onto a copper-copper sandwich can not reach both sides precisely at the very same time (as implied from our cooling rate measurements). The tentative transformation of our mono-jet into a double-jet variation (with a branched nozzle piece) did also not improve the results, when compared with a single propane-jet onto a copper-Thermanox sandwich.

    Notes: Abstract Tan-colored (tan) mutants, which retain the capacity to reduce acetylene, were selected and isolated fromRhizobium japonicum L-259 after growth in a glutamate-limited medium supplemented with tryptophan (trp). The mutants catabolized trp (500 mg per liter) to produce organe-colored fermentation broths containing extracellular indolepyruvic (IPA) and indoleacetic (IAA) acids. In contrast, parental strain L-259 produced a colorless broth containing only IAA when grown on the trp-supplemented medium. Trp alone was determined to be an incomplete nitrogen source for mutant growth and did not stimulate acetylenereduction activity.
