    Notes: Summary Amounts of chromosomal DNA were estimated for Feulgen-stained, ovarian cells from flies carrying certain mutant alleles of the otu (ovarian tumor) gene. Epithelial sheath cells and lumen cells were found to contain the diploid (2C) amount of DNA and therefore served as internal, cytophotometric standards. Mitotically active follicle cells over young tumors-from homozygous otu 1 females contained either the 2C or 4C amounts of DNA; whereas, the tumor cell population contained 2C, 4C and 8C nuclei and many intermediate values. Egg chambers also occur in homozygous otu 7 females. Follicle cells above these oocytes undergo a maximum of four cycles of endomitotic DNA replication. The accompanying nurse cells (PNC) contain polytene chromosomes. These undergo a maximum of 12 endonuclear replication cycles. The PNCs show the expected levels of DNA for the first 6 cycles and the fraction failing to replicate during subsequent cycles may be as small as 10%. Lower than expected levels of DNA were detected in PNCs from an otu 1/otu 3 ovary, reflecting roughly 20% underreplication. The latter PNCs may have been interrupted before DNA synthesis was concluded. No simple model of genomic underreplication accounts for the several different patterns of DNA behavior observed for various otu mutants.

    Notes: Abstract Amounts of Feulgen staining in individual sperm nuclei of Drosophila melanogaster were determined with a scanning microdensitometer and compared with the DNA-Feulgen levels found for hen erythrocyte nuclei, taken as a presumed cytophotometric standard of 2.5×10−12 g DNA per cell. Under these conditions, the amount of DNA estimated for the haploid male genome of Drosophila was about 0.18×10−12 g DNA, corresponding to a molecular weight of roughly 11× 1010 daltons. From a comparable series of measurements on Feulgen-stained nuclei of hemocytes and epithelial cells of the imaginal soma, values of approximately 0.35–0.36×10−12 g DNA were estimated for the diploid or 2C male genome of this species.

    Notes: Abstract Natural populations of triploid females resembling the gynogenetic teleost, Poecilia formosa (Girard), occur in northeastern Mexico where they intermingle with diploid populations of this species and the members of congeneric bisexual species such as P. mexicana or P. latipinna. Mitotic configurations from gill epithelial cells show 46 chromosomes for the diploid fishes, but 69 chromosomes for members of the triploid clones associated with P. formosa. Triploid females have erythrocytes that are significantly larger than those from diploid specimens and also show a roughly 50% elevation in the average DNA content of their somatic nuclei. Similar analyses of two functionally incompetent males of P. formosa, of a number of bisexual F1 and F2 hybrid offpsring from P. latipinna x P. mexicana, and of females from several other poeciliid species consistently show only diploid DNA levels and somatic chromosome complements where 22N=46. Demonstration of cytogenetic criteria by which females from triploid clones may be clearly distinguished from sympatric diploid specimens of P. formosa or P. mexicana leaves unresolved, for the present, problems of an appropriate systematic designation for natural populations of triploid gynogenetic fishes. The role of sympatric speciation in the evolution of poeciliid genomes is discussed in terms of alternative mechanisms to account for the persistence in nature of a vertebrate triploid of hybrid origin.

    Notes: Both two-wavelength microspectrophotometry of Feulgen-stained whole nuclei and autoradiography of H3-thymidine incorporation by giant salivary chromosomes in Drosophila virilis demonstrate a net decrease in the relative rate of salivary DNA synthesis during the late third instar and prepupal stages of development. Amounts of DNA-Feulgen per nucleus were distributed into several classes, the means of which closely approximated values projected as geometric multiples of the basic somatic DNA level estimated from hemocyte nuclei of the same larvae. Comparison of DNA polytene class frequencies showed no statistical difference between male larvae of different development stages, although female prepupae showed a greater frequency of nuclei in higher polytene classes when compared to male prepupae of the same age. Comparison of chromosomal H3-thymidine incorporation with previously described H3-histidine incorporation suggests that the amino acid labeling, which reaches a maximum during the prepupal period, has a physiological significance distinct from chromosomal endoreplication.
