    Description: We report on the isolation of Actinobacillus equuli ssp. haemolyticus from wound smears of a 2-year-old girl who was admitted to the hospital due to partial amputation of the distal phalanx of her right middle finger caused by a horse bite. A. equuli typically causes diseases in horses and only very few reports describing human infections (mostly associated with wounds) are available in the literature. Interestingly, although the bacteria could be found in consecutive samples taken at different points in time, there were no signs of advancing infection or inflammation. Moreover, the fingertip regenerated after 74 days under semi-occlusive dressings with very pleasant results. For strain identification two automated systems were employed producing discrepant results: VITEK 2 described the pathogens as Pasteurella pneumotropica while MALDI-TOF MS analysis revealed A. equuli . Sequence analysis of 16S rDNA gene finally confirmed A. equuli ssp. haemolyticus as the isolated strain. The antimicrobial susceptibility testing was performed according to the CLSI criteria for Pasteurella spp. Additionally we conducted a test according to the EUCAST criteria.
    Description: Wir berichten über den Nachweis von Actinobacillus equuli ssp. haemolyticus aus Wundabstrichen eines 2 Jahre alten Mädchens, welches sich wegen eines Pferdebisses in der Klinik vorstellte. Diese Verletzung führte zu einer teilweisen Amputation der distalen Phalanx ihres rechten Mittelfingers. Bei A. equuli handelt es sich um Bakterien, welche typischerweise Krankheiten bei Pferden verursachen. Bei Menschen findet A. equulii zumeist als Erreger von Wundinfektionen Erwähnung. Die Erreger konnten zu unterschiedlichen Zeitpunkten nachgewiesen werden. Während der gesamten Behandlungsdauer waren keine Zeichen einer fortschreitenden Infektion oder Entzündung zu beobachten. Die Fingerkuppe regenerierte nach 74 Tagen unter semi-occlusiven Verbänden mit einem sehr schönen Ergebnis. Die Erreger wurden mit Hilfe der beiden automatisierten Systeme VITEK 2 und MALDI-TOF MS identifiziert. Die biochemische Untersuchung mittels VITEK 2 identifizierte den Erreger als Pasteurella pneumotropica , wohingegen die massenspektrometrische Analyse A. equuli als Resultat lieferte. Die anschließende Sequenzierung des 16S rDNA Gens erbrachte das Ergebnis A. equuli ssp . haemolyticus und bestätigte somit die Identifikation der MALDI-TOF MS-Analyse. Wir erstellten für den Erreger Antibiogramme nach den Kriterien der amerikanischen Norm (CLSI) für Pasteurella spp . und stellten einen Vergleich zur europäischen Norm (EUCAST) an.

    Abstract: Among the Retroviridae, foamy viruses (FVs) exhibit an unusual way of particle assembly and a highly specific incorporation of envelope protein into progeny virions. We have analyzed deletions and point mutants of the prototypic FV gag gene for capsid assembly and egress, envelope protein incorporation, infectivity, and ultrastructure. Deletions introduced at the 3' end of gag revealed the first 297 amino acids (aa) to be sufficient for specific Env incorporation and export of particulate material. Deletions introduced at the 5' end showed the region between as 6 and 200 to be dispensable for virus capsid assembly but required for the incorporation of Env and particle egress. Point mutations were introduced in the 5' region of gag to target residues conserved among FVs from different species. Alanine substitutions of residues in a region between as 40 and 60 resulted in severe alterations in particle morphology. Furthermore, at position 50, this region harbors the conserved arginine that is presumably at the center of a signal sequence directing FV Gag proteins to a cytoplasmic assembly site

    Abstract: Azidothymidine (AZT) is a reverse transcriptase (RT) inhibitor that efficiently blocks the replication of spumaretroviruses or foamy viruses (FVs). To more precisely elucidate the mechanism of action of the FV RT enzyme, we generated an AZT-resistant FV in cell culture. Biologically resistant virus was obtained for simian foamy virus from macaque (SFVmac), which was insensitive to AZT concentrations of 1 mM, but not for FVs derived from chimpanzees. Nucleotide sequencing revealed four non-silent mutations in the pol gene. Introduction of these mutations into an infectious molecular clone identified all changes to be required for the fully AZT-resistant phenotype of SFVmac. The alteration of individual sites showed that AZT resistance in SFVmac was likely acquired by consecutive acquisition of pol mutations in a defined order, because some alterations on their own did not result in an efficiently replicating virus, neither in the presence nor in the absence of AZT. The introduction of the mutations into the RT of the closely related prototypic FV (PFV) did not yield an AZT-resistant virus, instead they significantly impaired the viral fitness.     Notes: Abstract Serum bile acids were fractionated by high-performance liquid chromatography (HPLC) in 13 control and 8 cases of liver disease in horses. The severity and type of liver injury was determined by histopathological examination of biopsy and/or necropsy specimens. The total serum bile acids (tSBA) were determined in these horses by an enzymatic method (SBA-EA) and by summation of the bile acids (SBA-LC) as fractionated by the HPLC. The SBA-LC were generally higher than the SBA-EA in both the controls and liver disease and they did not parallel each other. The primary bile acids, total cholates and total chenodeoxycholates accounted for most of the tSBA increases in liver disease. There was a shift in profile from taurocholate to free (unconjugated) cholate in direct relation to the severity of the liver injury. Among the secondary bile acids, total deoxycholates and total taurodeoxycholates increased at random. The pattern of the SBA profile in relation to the severity of the liver disease suggested that hepatocellular excretion is the most sensitive step in the enterohepatic circulation of the bile acids.

    Notes: Zusammenfassung Zusammenfassend kann festgestellt werden, daß die Prognose des offenen Bisses beim Sigmatismus interdentalis nur günstig sein kann, wenn 1. die Behandlung der Anomalie vom Kieferorthopäden und Sprachphysiologen gemeinsam durchgeführt wird und 2. die den Phänotypus bestimmenden Faktoren, wie Alter, Reaktionslage, Konstitution des Patienten, Größe der Anomalie, Sprachlautbildung (Zungenfunktion) usw. mit der Wahl der therapeutischen Hilfsmittel abgestimmt sind. 3. Der geeignetste Augenblick zum Eingreifen besteht naturgemäß während des Schneidezahnwechsels, denn von hier ab bis zur geschlechtlichen Reife — dem 12. bis 14. Lebensjahr — kann man das noch in vollem Fluß befindliche Wachstum des Gebisses mit Hilfe funktionskieferorthopädischer Apparate, zu denen die elastischen Gebißformer nach Bimler—für den offenen Biß die modifizierte Form—gehören, dergestalt beeinflussen, daß nur den von uns gewünschten Erbanlagen günstige äußere Bedingungen geschaffen werden; alle störenden Umwelteinflüsse aber von den weniger durchschlagskräftigen Erbanlagen ferngehalten werden.

    Notes: Zusammenfassung 1. Eine Unterentwicklung des Oberkiefers läßt sich nur durch kieferbezügliche Meßmethoden diagnostizieren. 2. Es wird eine Meßmethode bekanntgegeben, die bestimmte Relationen zwischen der transversal-sagittalen Ausdehnung des Zahnbogens und der apikalen Basis berücksichtigt, welche auf Grund der individuellen SI-Summe errechnet und klinisch erprobt wurde. 3. Daraus entstand eine Tabelle, die für jede SI-Summe die sagittale Oberkieferwurzelgrundebene angibt. 4. An Stelle der nicht angelegten mittleren und seitlichen Oberkieferschneidezähne kann die Berechnung der sagittalen Oberkieferwurzelgrundebene von den Unterkieferschneidezähnen vorgenommen werden. Als vorderer Meßpunkt für die sagittale Oberkieferwurzelgrundebene treten die Eckzähne an Stelle der mittleren oberen Inzisivi. Schrifttumsverzeichnis beim Verfasser.
