    Notes: Summary With growing cells of a Nocardia sp., isolated from soil, the degradation of 5-pregnene-3β, 20β-diol into 3-[5-oxo-7aβ-methyl-1β (1′β-hydroxo)-ethyl-3aα-perhydroindane-4α]-propionic acid was investigated. The results show that iron is essential for production of the perhydroindanpropionic acid, that this production is greatly enhanced by the presence of calcium and that it is maximal in the pH range 7.0–7.5.

    Notes: Abstract Hexose phosphate synthetase activity was found in cell-free extracts of methanol-grown Candida boidinii. Incubation of this crude extract with 14C-formaldehyde and D-ribose-5-phosphate leads to incorporation of radioactivity into fructose-and glucose phosphates. Cells grown on glucose lack the hexose phosphate synthetase activity. No hydroxypyruvate reductase activity, the key enzyme of the serine pathway was found. These results indicate that during growth of C. boidinii on methanol, cell constituents are made by a sugar phosphate pathway similar in concept, if not in absolute molecular detail, to the ribose phosphate cycle in C1-metabolizing bacteria.

    Notes: Summary The effect of several parameters (temperature; pH; main carbon source; time and amount of β-sitosterol addition; Tween 20, 40, 60, 80; Span 20; pluronic F 68, L 64) on the conversion of β-sitosterol to 3-(5α-hydroxy-7aβ-methyl-1-oxo-3aα-H-hexahydroindan-4α-yl) propionic acid (I) by Nocardia sp. M. 29–40 was investigated. A maximal theoretical yield of 65 mol% I (with respect to substrate added) could be achieved during cultivation at pH 8.0 in presence of 6 g/l Tween 40 or Tween 60. Tween 40 and Tween 60 stimulate β-sitosterol cooxidation not by improving the substrate suspension but by providing a fatty acid component as precursor for biosynthesis of surface active cell wall lipids.

    Notes: Summary A bacterium capable of growth on methanol and some organic acids as sole source of carbon and energy has been isolated and designated Pseudomonas 3ab. This facultative methylotrophic organism apparently utilizes the serine pathway of formaldehyde fixation. When methanol was used as the sole carbon source for growth, L-serine production by Pseudomonas 3ab occurred upon the addition of glycine and methanol at the end of the exponential growth phase. The maximum yield of L-serine (4.7 g/l) was obtained when 20 g/l glycine and 8 g/l methanol were added and the pH of the culture medium was changed to 8.5. Although Pseudomonas 3ab is unable to grow on L-serine or glycine, it is very active in decomposing these amino acids. The degradation of L-serine and glycine has been shown to be pH-dependent with a minimum at pH 8.5–9.0.

    Notes: Summary Growing and resting cells of the yeast Trigonopsis variabilis (DSM 70714) can be used for the enantioselective reduction of the organosilicon compound acetyldimethylphenylsilane (1) to give optically active (R)-(1-hydroxyethyl)dimethylphenylsilane [(R)-2] in good yields. The enantiomeric purity of the isolated product was determined to be 62–86% ee depending on the substrate concentration used. Both substrate and product caused an inhibition of the reaction at concentrations higher than 0.35 and 0.5 g/l, respectively. Besides, higher substrate and product concentrations led to increased formation of the by-product 1,1,3,3-tetramethyl-1,3-diphenyldisiloxane. Considering the limiting substrate and product concentrations, it was possible to use the same biomass at least 5 times without significant loss of enzyme activity. 3-Methyl-3-phenyl-2-butanone (5) and acetyldimethylphenylgermane (7), which represent carbon and germanium analogues of 1, were also found to be accepted as substrates by Trigonopsis variabilis (DSM 70714). The reduction rates of the silicon (1) and germanium compound (7) were much higher than the transformation rate of the corresponding carbon analogue 5.

    Notes: Summary rac-2-Acetoxy-1,1-dimethyl-1-sila-cyclohexane (rac-2) was synthesized by esterification of rac-1,1-dimethyl-1-sila-cyclohexan-2-ol (rac-1) with acetic anhydride. Enantioselective hydrolysis of rac-2 in aqueous solution, catalysed by a crude lipase preparation of Candida cylindracea (EC 3.1.1.3), led to the formation of (S)-1 (95% ee). Enantioselective transesterification of rac-1 with triacetin in isooctane, catalysed by the same enzyme preparation, yielded (S)-2 (95% ee), which was separated by chromatography from non-reacted (R)-1 (96% ee). Recrystallization led to an improvement of the enantiomeric purity of (R)-1 and (S)-1 up to 〉98% ee. Thus the enantiomers of rac-1 were prepared (100 mg scale) with high enantiomeric purities by the use of two different types of enzyme-catalysed reaction.

    Notes: Summary During screening for biosurfactants among marine, n-alkane-utilizing bacteria, low- and high-molecular surface-active substances were detected. The marine bacterial strain MM1 was found to synthesize a novel glycolipid that has not so far been cited in the literature. Both 1H, 13C-nuclear magnetic resonance spectroscopic and positive ion fast atom bombardment mass spectrometer studies led to the identification of a glucose lipid consisting of four 3-OH-decanoic acids, which are linked together by ester bonds. The lipophilic moiety is coupled glycosidically with C-1 of glucose. The glucolipid reduced the surface tension from 72 mN/m to 30 mN/m while the minimum interfacial tension towards n-hexadecane was lowered to values smaller than 5 mN/m.

    Description / Table of Contents: Zusammenfassung Aus einer Bodenprobe wurde eine Hefe, Candida boidinii, isoliert, welche auf Methanol als einziger C-Quelle wachsen kann. Als Wachstumsfaktor benötigt dieser Hefestamm Biotin in sehr geringer Konzentration. Unter günstigen Kulturbedingungen beträgt die Zellausbeute pro 1000 ml Kulturmedium 2,3g Trockenmasse bei Zugabe von 1% (v/v) Methanol, und 8,3g bei 4% Methanol. Das Wachstum wird bei Zusatz von 5% Methanol zum Minimalmedium vollständig gehemmt. Der Stamm verwertet Kohlenhydrate und Äthanol schneller als Methanol oder Milchsäure. Die Enzyme für den Methanol-Stoffwechsel scheinen jedoch konstitutiv zu sein. Die optimalen Kulturbedingungen für kurze Generationszeiten und hohe Zellausbeuten auf Methanol sind: 28°C, NH4 + als Stickstoffquelle und pH 5,0. Die Elementaranalyse ergab folgende Werte für die Zusammensetzung der Hefezellen: 42,81% C, 7,23% H und 5,54% N. Die Aminosäuren (in den Zellen) wurden mit dem Aminosäure-Analysator quantitativ bestimmt.
    Notes: Summary A yeast, Candida boidinii, isolated from soil, capable of growing on a medium containing methanol as the only carbon source is described. Biotin is required in very low concentration as a growth factor. In a study on the effect of the methanol concentration on the cell growth under favorable conditions, the cell yield was 2.3 g (dry weight) with 1% (v/v) methanol and 8.3 g with 4% methanol per 1000 ml of culture medium. However, the growth was inhibited by 5% methanol. The strain assimilated carbohydrate and ethanol faster than methanol or lactate. The enzymes for the methanol metabolism are probably constitutive. Optimal conditions for rapid growth and high cell yeild from methanol were found to be: 28°C, NH4 + as nitrogen source and pH 5.0. The cell composition was as follows: 42.81% C, 7.23% H and 5.54% N. Amino acids in the cells were analyzed by the amino acid autoanalyzer.

A specific NAD-dependent formaldehyde dehydrogenase requiring reduced glutathione has been partially purified. Furthermore, a NAD-linked formate dehydrogenase was found in cell-free extracts. The synthesis of these two enzymes is induced by methanol and repressed by glucose. The possible significance of these enzymes in the energy-generating system is discussed.
