    Abstract: Dot1l encodes histone H3 K79 methyltransferase Dot1a. Mice with Dot1l deficiency in renal Aqp2-expressing cells (Dot1l(AC)) develop polyuria by unknown mechanisms. Here, we report that Aqp5 links Dot1l deletion to polyuria through Aqp2. cDNA array analysis revealed and real-time RT-qPCR validated Aqp5 as the most upregulated gene in Dot1l(AC) vs. control mice. Aqp5 protein is barely detectable in controls, but robustly expressed in the Dot1l(AC) kidneys, where it colocalizes with Aqp2. The upregulation of Aqp5 is coupled with reduced association of Dot1a and H3 dimethyl K79 with specific subregions in Aqp5 5' flanking region in Dot1l(AC) vs. control mice. In vitro studies in IMCD3, MLE-15 and 293Tcells using multiple approaches including real-time RT-qPCR, luciferase reporter assay, cell surface biotinylation assay, colocalization, and co-immunoprecipitation uncovered that Dot1a represses Aqp5. Human AQP5 interacts with AQP2 and impairs its cell surface localization. The AQP5/AQP2 complex partially resides in the ER/Golgi. Consistently, AQP5 is expressed in none of 15 normal controls, but in all of 17 kidney biopsies from patients with diabetic nephropathy. In the patients with diabetic nephropathy, AQP5 colocalizes with AQP2 in the perinuclear region and AQP5 expression is associated with impaired cellular H3 dimethyl K79. Taken together, these data for the first time identify Aqp5 as a Dot1a potential transcriptional target, and an Aqp2 binding partner and regulator, and suggest that the upregulated Aqp5 may contribute to polyuria, possibly by impairing Aqp2 membrane localization, in Dot1l(AC) mice and in patients with diabetic nephropathy.

    Notes: Abstract The changes in the neurofilament medium and high molecular weight subunits (NF150 and NF200) in the developing and transected spinal cords of bullfrog tadpoles were studied. A monoclonal antibody recognizing the nonphosphorylated epitope of NF150, NF150D, stained the neuronal cell bodies and axons, whereas other antibodies against the phosphorylated NFs, NF150P or NF200P, labeled chiefly the axons. During development, the intensity of axonal staining by the anti-NF150D in the ventral fasciculi in younger tadpoles appeared stronger than older animals, but the reverse was seen for NF150P and NF200P. Complete signal transection of stage IV tadpoles resulted in degeneration and then regeneration of the cord tissue of both cut ends. Each stump lengthened by about 350 μm in the 4 weeks after the lesion. In the proximal stumps, the levels of NF150P or NF200P in the ventral axons at 550-350 μm proximal to the transection site increased notably by about 24–73% of the control value 7–28 days post-transection; however, the content of NF150D was decreased. The densities of NF150D and NF150P protein spots on the Coomassie blue-stained two-dimensional gels of the normal and injured cords also displayed alterations similar to the immunocytochemical data. Intense labeling by the anti-NF150P or NF200P was present in the cell bodies of axotomized motor neurons in the ventral horn. The results suggest that central axonal regeneration may be accompanied by upregulated phosphorylated neurofilament proteins.

    Notes: Abstract  Thirty-four cases of primary non-Hodgkin’s lymphoma of the oral cavity were investigated for their clinical findings, histopathological features, immunophenotypes and association with Epstein-Barr virus (EBV). Four cases (12%) were natural killer/T-cell lymphomas, 3 (9%) were T-cell lymphomas and 27 (79%) were B-cell lymphomas. Compared with T- and B-cell lymphomas, NK/T-cell lymphomas had a male predominance (M:F 4:0), and most presented as ulceration of the palate and/or maxillary gingiva. Histologically, the lesions showed diffuse infiltration of medium-sized or large lymphoid tumour cells. Angiocentricity and/or angioinvasion were found in all 4 cases. The immunophenotypes of the NK/T-cell lymphomas were CD3+, CD43+, CD45RO+, CD56+ and TIA-1+. EBV was detected in 2 NK/T-cell lymphomas by in situ hybridization (ISH) and polymerase chain reaction (PCR) methods, and was not detected in T- and B-cell lymphomas. The survival rate of patients with NK/T-cell lymphoma was zero, but the survival rates for patients with T-cell and B-cell lymphomas were 67% and 38%, respectively. It appears that NK/T-cell lymphomas of the oral cavity have a predilection for originating in the palate and maxillary gingiva and are aggressive neoplasms. EBV positivity might be associated with more aggressive behaviour.

    Notes: 4 O7 crystal is a new nonlinear optical crystal. For the first time the anisotropy of the velocities of its longitudinal and surface acoustic waves (SAWs) are determined by laser ultrasonic technique. The velocities of surface waves for X-, Y-, and Z-cut crystals are also calculated. The theoretical calculations of slowness curves are in good agreement with experimental results. The SAW slowness curve is elliptical for Y- or Z-cut crystal wafers, and circular for an X-cut wafer.

    Notes: Abstract.  Consumption of trichlorfon-poisoned fish by women in a small Hungarian village has been associated with trisomy resulting from an error of meiosis II in oogenesis. We therefore examined mouse oocytes exposed for 3 h during fertilization to 50 µg/ml trichlorfon. Spindle morphology was not visibly altered by the pesticide. Chromosomes segregated normally at anaphase II with no induction of aneuploidy. However, formation of a spindle was disturbed in many oocytes resuming meiosis I in the presence of trichlorfon. In spite of the spindle aberrations and the failure of bivalents to align properly at the equator, oocytes did not become meiotically arrested but progressed to metaphase II. At this stage, spindles were highly abnormal, and chromosomes were often totally unaligned, unattached or dispersed on the elongated and disorganized spindle. By causing spindle aberrations and influencing chromosome congression, trichlorfon appears, therefore, to predispose mammalian oocytes to random chromosome segregation, especially when they undergo a first division and develop to metaphase II during exposure. This is the first case in which environmentally induced human trisomy can be correlated with spindle aberrations induced by chemical exposure. Our observations suggest that oocytes may not possess a checkpoint sensing displacement of chromosomes from the equator at meiosis I and may therefore be prone to nondisjunction.

    Notes: Summary Uncoated fused-silica capillary tubing has found widespread use in GC and GC/MS, e. g., in splitless and oncolumn injection, in the open-split interface used in GC/MS, and in hybrid fused-silica/glass capillary columns. As the uncoated capillary is usually more active than the coated one, it is very important to pay sufficient attention to the deactivation of the uncoated capillary. Otherwise the uncoated portion used as column inlet and/or outlet may become the main source of column activity and of analytical error. This case is especially serious under temperature-programmed runs. The effect of film thickness and temperature program on column activity is also discussed.
