    Notes: We have studied the effect of a single injection of reserpine (5 mg/kg, s.c.) on the synaptic vesicle monoamine transporter (VMAT) density in the rat striatum, using two labelling procedures: radioimmunolabelling with an antibody against VMAT, and binding of the specific ligand [3H]dihydrotetrabenazine ([3H]TBZOH). In the rostral and medial striatum, the distribution of VMAT immunoreactivity displayed the highest density in the lateral subregions. In the caudal part of the striatum, VMAT immunoreactivity showed increasing density from dorsal to ventral subregions. The VMAT immunoreactivity was not altered 2 and 30 days after the reserpine injection, whereas [3H]TBZOH binding site density, measured on adjacent slices, showed a dramatic decrease at day 2 and a moderate recovery at day 30, suggesting that despite a persistent blockade of [3H]TBZOH binding sites, VMAT protein density was unchanged.

    Notes: Abstract We evaluated a new sample processor, the PULSIFIER®, for preparing food suspensions for microbiological analysis. In a comparative study on 96 samples of representative foods, Total Aerobic Counts (TACs) from “pulsified'’samples equaled or exceeded those from a Stomacher® 400 at levels up to 107 CFU/g, ranging up to 2x higher at low count levels. Except for already comminuted products, pulsified suspensions always contained much less food debris. For example, for 8 replicate samples of celery and carrot, average TACs of pulsified samples were higher (1.3x and 2.5x, respectively) although pulsified suspensions were clear while stomachered suspensions contained enough debris to interfere with pipetting. The superior quality of microbial suspensions from the Pulsifier has implications for general analysis as well as techniques such as PCR and ATP bioluminescence, which are easily perturbed by tissue extracts.

    Notes: Time-dependent phototropism (TDP), sometimes called second positive curvature, occurs when the duration of phototropic stimulation with blue light (B) exceeds a few minutes. TDP was characterized in maize (Zea mays L.) coleoptiles raised under continuous red light (R). Subsequently, coleoptiles adapted to darkness were used to investigate the effect of R on TDP. It was found that TDP, which is induced in R-grown coleoptiles, does not occur in dark-adapted coleoptiles and that dark-adapted coleoptiles begin to show TDP after treatment with R. The TDP responsiveness became maximal 1–2 h after treatment with a R pulse and decreased during the next few hours. At least 10 min was required after a short pulse of R before the coleoptile began to respond to B for the induction of TDP. The effect of R in establishing the TDP responsiveness was totally suppressed by a pulse of far-red light given immediately after an inductive pulse of R. It is concluded that the mechanism of TDP requires for its establishment a R signal perceived by phytochrome. The TDP of R-grown and R-pretreated coleoptiles showed relationships to stimulation times and fluence rates that are similar to those reported for oat coleoptiles, except that TDP of maize showed a sharp increase in its magnitude within a narrow range of stimulation times as short as 5–10 min.
