    Notes: Abstract: New band structures have been observed in 112Pd, 113Pd, 115Pd and 118Pd produced via the fusion-fission reaction 12C+238U at a bombarding energy of 90 MeV. γ-γ-γ coincidence measurements have been performed with the Euroball III array. The first crossings observed in even-mass palladium up to A=118 are interpreted as the alignment of a νh 11/2 pair. This interpretation is supported by the rotational behavior of odd-mass palladium. Hartree-Fock-Bogoliubov calculations using D1S-Gogny force suggest a similar interpretation consistent with a prolate deformation.

    Description / Table of Contents: Multimodal therapy of oral squamous cell carcinomas today is based on surgery, radiotherapy and chemotherapy. Despite the combination of all three therapeutic options, there is still a large number of treatment failures and therefore major questions remain. Recent investigations suggest that mutations of the p53 tumor supressor gene may account for some of the therapeutic failures. Inactivation of the gene may be an important determinant of the efficiacy of today’s multimodal therapy protocols. In 90 patients with squamous cell carcinomas of the oral cavity biopsy specimens were taken before and after preoperative radiochemotherapy. From all patients, biopsy and resection material was available for immunohistochemical analysis of p53. After radiation treatment, 51 patients (57%) showed a complete response; 39 patients (43%) only showed a partial response or did not respond at all. Among the responders, 82% of the pretherapeutic tumors were p53 positive, whereas among the nonresponders only 56% of the pretherapeutic tumors were p53 positive. The majority of the residual tumors were also p53 negative according to immunohistology after radiation treatment. In our study, detection of p53 protein by immunohistochemistry seemed to be connected with a more radiosensitive reaction of the tumors. Nevertheless, successful strategies for radiation therapy may need to take into account the tissue of origin and the status of p53 in the tumor.
    Notes: Trotz der seit einigen Jahrzehnten in der Behandlung der Mundhöhlenkarzinome eingesetzten Kombinationstherapie ist die Rezidivrate unverändert hoch. Jüngsten Untersuchungen an Plattenepithelkarzinomzellinien zufolge scheinen Mutationen des p53-Tumorsuppressorgens für einen Teil der Therapieresistenzen insbesondere in der Radiotherapie mitverantwortlich zu sein. In einer retrospektiven Studie wurde bei 90 Patienten mit Plattenepithelkarzinomen der Mundhöhle, die mit einer präoperativen Radiochemotherapie und nachfolgender Tumorresektion behandelt wurden, die p53-Exrpession vor und nach der Radiochemotherapie immunhistologisch untersucht und mit dem Ansprechen auf die Therapie korreliert. Nach der Radiochemotherapie waren bei 51 (57%) Patienten klinisch und histopathologisch im Operationsresektat keine Tumoranteile mehr nachweisbar. 39 (43%) Patienten zeigten keine oder nur eine geringe Reaktion auf die Therapie. Die therapiesensitiven Tumoren wiesen in den prätherapeutischen Biopsien in 82% der Fälle eine Expression des p53 auf, die therapieresistenten Tumoren lediglich in 56%. Nach Abschluß der Radiatio konnte nur noch in 4 der 39 Resttumoren p53 nachgewiesen werden. In unserer Untersuchung schien die Expression des p53-Gens mit einer erhöhten Therapiesensibilität einherzugehen.

    Notes: Abstract: Isomeric states have been observed in fission-fragments produced by spontaneous fission of 252Cf. These states are found in neutron rich nuclei of different structure and deformations. About 50 isomeric nuclei have been observed using coincidences between γ-rays identified in EUROGAM II and fission fragments detected in photovoltaic cells (SAPhIR). Lifetimes in the range from 20 ns to 2μs have been measured. Presented calculations based on HFB +D1S force on new measured isomeric states in the 152,154,156Nd show evidence for K-isomers.

    Notes: Abstract Genomic and cDNA clones derived from the gene (dodA) coding for DOPA dioxygenase, a key enzyme in the betalain pathway, were obtained from the basidiomycete Amanita muscaria. A cDNA library was established in the phage λZapII and dodA clones were isolated using polyclonal antibodies raised against the purified enzyme. Their identity was confirmed by comparison of the deduced amino acid sequence with the sequence of several tryptic peptide fragments of DOPA dioxygenase. The gene coded for a 228-amino acid protein that showed no homology to published sequences. The coding region was interrupted by five short introns. Regulation was shown to occur at the transcriptional level; the mRNA accumulated to high levels only in the coloured cap tissue. dodA was found to be a single-copy gene in A. muscaria. To our knowledge, this is the first gene from the betalain pathway to be cloned. It encodes a type of aromatic ring-cleaving dioxygenase that has not been previously described.

    Notes: Abstract Cranio-spinal irradiation is the gold standard treatment used in non metastatic medulloblastoma as prophylaxis against central nervous system (CNS) metastases. However, given the severe late effects caused by this procedure in children under 3 years of age, most pediatric oncologists are currently treating these patients with conventional chemotherapy in order to postpone or even avoid irradiation. In the French Society of Pediatric Oncology ('FOP) this attitude has been adopted since 1987. Among the patients treated without radiotherapy, 20 relapsed while on conventional chemotherapy and were entered in a study of high-dose chemotherapy (HDC) followed by ABMT. Their median age at diagnosis was 23 months (R5-71) and the relapse occurred at a median time of 6.3 months after the initiation of chemotherapy. Complete surgical removal of the local relapse was the first treatment in 4/20 patients who were not evaluable for response. Sixteen of the twenty patients had measurable disease at the primary site (9 patients), or at metastatic sites (3 patients) or both (4 patients). The conditioning regimen consisted of combination Busulfan 600 mg/m2 over 4 days and Thiotepa 900 mg/m2 over three days. After recovery from aplasia, patients with a local relapse received local radiotherapy limited to posterior fossa. Results. Among the 16 patients with measurable disease, 6 CR, 6 PR, 3 NR, were observed following HDC (response rate 75 %). One patient was not evaluable. For the 20 patients, the EFS is 50%. Among the surviving patients, the median follow up is 31 months post BMT (R12-82). Ten patients who developped a local relapse or local progression are alive with NED without craniospinal irradiation. Among the 7 patients who developped metastases or progression of metastases, only one is alive. Toxicity was high but manageable: the median duration of granulocytopenia 〈 0.5 × 109/1 and thrombocytopenia 〈 50 × 109/1 was 13 and 41 days respectively. Bacteremia was documented in 4 cases. Grade 〉 2 mucositis and diarrhea were observed in 60% of patients. One complication-related death occurred 1 month post BMT. Conclusion. With a 75 % response rate, this HDC proved to be very efficient in relapsed medulloblastoma. A longer follow up is necessary to demonstrate whether, after a local relapse, HDC could replace craniospinal irradiation as prophylaxis against CNS metastases.

    Notes: Abstract As initial steps to define how the 26S proteasome degrades ubiquitinated proteins in plants, we have characterized many of the subunits that comprise the proteolytic complex from Arabidopsis thaliana. A set of 23 Arabidopsis genes encoding the full complement of core particle (CP) subunits and a collection encoding 12 out of 18 known eukaryotic regulatory particle (RP) subunits, including six AAA-ATPase subunits, were identified. Several of these 26S proteasome genes could complement yeast strains missing the corresponding orthologs. Using this ability of plant subunits to functionally replace yeast counterparts, a parallel structure/function analysis was performed with the RP subunit RPN10/MCB1, a putative receptor for ubiquitin conjugates. RPN10 is not essential for yeast viability but is required for amino acid analog tolerance and degradation of proteins via the ubiquitin-fusion degradation pathway, a subpathway within the ubiquitin system. Surprisingly, we found that the C-terminal motif required for conjugate recognition by RPN10 is not essential for in vivo functions. Instead, a domain near the N-terminus is required. We have begun to exploit the moss Physcomitrella patens as a model to characterize the plant 26S proteasome using reverse genetics. By homologous recombination, we have successfully disrupted the RPN10 gene. Unlike yeast rpn10Δ strains which grow normally, Physcomitrella rpn10Δ strains are developmentally arrested, being unable to initiate gametophorogenesis. Further analysis of these mutants revealed that RPN10 is likely required for a developmental program triggered by plant hormones.
