    Description / Table of Contents: Summary Background and Objective: Pemphigoid gestationis (PG) is a rare pregnancy-associated autoimmune bullous disease characterized by autoantibodies to the 180 kD bullous pemphigoid antigen (BP180). The clinical spectrum of PG is polymorphic and for diagnostic purposes, a skin biopsy is usually taken demonstrating the deposition of autoantibodies. Patients and Methods: From 2 patients, skin biopsies were obtained for histopathologic and immunofluorescence studies. Circulating autoantibodies were characterized by immunoblotting and ELISA using a recombinant form of the immunodominant BP180 NC16 A domain. Results: The 2 PG patients described here did not show blisters but complained about severe itching. In the first case, PG presented in the first trimester of the second pregnancy as an erythema-multiforme-like disease. The second patient developed urticarial plaques a few days after delivery. PG was diagnosed by the detection of autoantibodies against recombinant BP180 NC16 A by immunoblot and ELISA analysis and confirmed by linear deposits of C3 at the cutaneous basement membrane zone on direct immunofluorecence microscopy. Skin lesions healed with oral prednisolon. Conclusions: In our two patients, non-bullous PG could be diagnosed by serological tests. Immunmoblotting and ELISA might be sensitive and specific tools when screening sera of patients with pruritic skin lesions in pregnancy for the presence of autoantibodies to BP180. In some cases, these newer techniques may make a skin biopsy unnecessary.
    Notes: Zusammenfassung Hintergrund und Fragestellung: Das Pemphigoid gestationis (PG) ist eine seltene, schwangerschaftsspezifische Autoimmundermatose mit Autoantikörpern gegen das 180 kD bullöse Pemphigoid Antigen (BP180). Das klinische Bild ist polymorph. Die Diagnose des PG wird meist anhand einer Biopsie mit Nachweis der Autoantikörper in der Haut der Patientinnen gesichert. Patienten/Methodik: Bei 2 Patientinnen wurden Hautbiopsien histopatholgisch und immunfluoreszenzoptisch untersucht. Die Autoantikörper wurden mittels Immunoblot und ELISA unter Verwendung einer rekombinanten Form der immundominanten BP180-NC16-A-Domäne charakterisiert. Ergebnisse: Bei den beiden vorgestellten Patientinnen verlief das PG ohne Blasenbildung, jedoch mit starkem Juckreiz. Bei unserer ersten Patientin manifestierte sich die Erkrankung im 1. Trimenon unter dem Bild eines Erythema multiforme, der 2. Fall trat wenige Tage postpartal unter dem Bild einer Urtikaria auf. Die Diagnose wurde jeweils durch den Nachweis von Antikörpern gegen rekombinantes BP180 NC16 A im Immunoblot und ELISA und durch lineare C3-Ablagerungen an der Basalmembran in der direkten Immunfluoreszenz gesichert. Die Hautveränderungen heilten bei beiden Patientinnen unter oraler Prednisolontherapie ab. Schlussfolgerung: Bei den beiden vorgestellten Patientinnen wurde anhand von Serumuntersuchungen ein PG ohne Blasenbildung diagnostiziert. Sowohl Immunoblot als auch ELISA erscheinen aufgrund ihrer hohen Sensitivität und Spezifität als Screeningverfahren zum Nachweis von Autoantikörpern gegen BP180 bei pruriginösen Schwangerschaftsdermatosen geeignet. In der Zukunft dürften sie mitunter die bisher unverzichtbare Hautbiopsie zur Diagnose eines PG ersetzen.

    Notes: Abstract Resonance ionization mass spectrometry (RIMS) has been tested for the isotope-selective determination of trace amounts of plutonium. An atomic beam is formed by evaporating plutonium atoms from a rhenium-filament heated to 1800 °C. The radiation of a pulsed dye laser excites the atoms in a two-photon process (λ=595.2 nm) followed by photoionization of the excited atoms. Mass selectivity is obtained by use of a time-of-flight spectrometer. A resonant signal of239Pu was measured with 1013 atoms deposited on the filament.

    Notes: Abstract Laser photoionization has been used to detect trace amounts of plutonium. A high sensitivity and selectivity has been achieved by applying three-step excitation and ionization of the plutonium atoms with high pulse-repetition rates and additional mass determination by time-of-flight measurements. A laser system was developed which consists of a copper vapor laser pumping three dye lasers simultaneously. Samples containing between 1010 and 1012 atoms of239Pu on Re filaments were measured yielding strong resonance signals with maximum ion count rates of several kHz at a vanishingly low background. A detection efficiency of 10−7 was determined allowing the detection of about 108 plutonium atoms in a sample.

    Notes: Abstract The neutron-rich isotope154Nd has been separated from235U fission products with the HELIum-jet On-line mass Separator HELIOS and investigated by γ-singles, γγ- and γX-coincidence measurements. The Z- and A-assignment have been confirmed by the observation of promethium KX-rays and by the genetic relationship to the daughter product 1.7-min154Pm. A half-life of 26±2 s has been obtained for154Nd, and a partial decay scheme is proposed. A 40 s activity previously assigned to154Nd could not be confirmed.

    Notes: Abstract Integral fission cross sections in the system238U+238U were measured at beam energies below the interaction barrierV C. Scattering angle dependent probabilities and integral cross sections for Coulomb fission were calculated. It is concluded that earlier observed discrepancies between measured and calculated angular distributions for the one-neutron transfer product239U cannot be explained by sequential fission. Multi-nucleon transfer induced fission is observed down to energies (0.90±0.02)×VC.

    Notes: Summary Glucagon-like peptide-1(7–36)amide [GLP-1(7–36)amide] is supposed to be an important physiologic incretin. Recently, high affinity receptors for GLP-1(7–36)amide have been demonstrated on rat insulinomaderived RINm5F cells. The present study examined the internalization and degradation of the GLP-1-receptor complex. Internalization of the peptide was time- and temperature-dependent. At 37°C binding and internalization was rapid. At 60 min 35% of125I-labeled GLP-1(7–36)amide was internalized. Incubation in the presence of increasing concentrations of nonlabeled GLP-1(7–36)amide resulted in a decrease of internalization of125I-labeled peptide indicating that this process is saturable. Incubation in the presence of 0.2 mM chloroquine, an inhibitor of intracellular hormone degradation, resulted in intracellular accumulation of125I-GLP-1(7–36)amide. HPLC-supported analysis of cell content after internalization of125I-GLP-1(7–36)amide during a 60-min incubation period at 37°C revealed an elution profile showing two maxima of radioactivity: one represented intact labeled GLP-1(7–36)amide, the other an intracellular degradation product of the peptide. Chloroquine caused a 5-fold increase of the peak representing intact125I-GLP-1(7–36)amide thus demonstrating inhibition of degradation of labelled peptide. Furthermore, a 4-fold increase of the other peak occurred possibly mirroring a delay of release of degradation products by chloroquine. It was excluded that chloroquine is able to interfere with GLP-1(7–36)amidebinding to its receptor.
