    Notes: Abstract The fluorescence depolarization of 1,6-diphenyl-hexatriene was used to study the dynamic properties of the hydrophobic regions of the lipid envelopes of ortho- and paramyxoviruses as well as of the Rous sarcoma virus and of the membrane lipids of susceptible and nonsusceptible cells. The systems investigated where active and inactive influenza viruses, and NDV virus acting on chick embryo fibroblasts and Rous sarcoma virus acting on susceptible (C/E) and nonsusceptible (C/B) chicken-cell. Polarization degrees and mean rotational correlation times of DPH embedded in viral lipids were significantly higher than those of DPH in the cell membranes, due to a higher rigidity of the virus envelopes. When suspensions of labelled viruses and unlabelled cells or unlabelled viruses and labelled cells were mixed, a characteristic change of the fluorescence polarization degrees with time was observed. This behaviour was ascribed to a label transfer from virus to cell membranes or vice versa. While the rate constants of label transfer from virus to cells and cells to virus were about the same for the penetrating viruses the rate constants of label release from inactive virus to cells were much larger than for the migration in the opposite direction.

    Notes: Abstract We recently presented evidence showing that the visible CD spectrum of the purple membrane from Halobacterium halobium consists of two contributions: a broad positive band centered at the absorption maximum due to the interaction of the chromophore with the protein to which it is bound, and an exciton coupling band due to the interaction between chromophores of adjacent bacteriorhodopsin molecules in the hexagonal surface lattice (Heyn et al., 1975). This interpretation receives strong support from the present experiments in which the chromophore-free membrane is reconstituted by the addition of retinal. Since the coupling signal arises from the interaction between pairs of neighboring chromophores, its contribution to the spectrum would be expected to be very small in the initial stages of the titration experiment, but increasing quadratically with the percentage reconstitution. The broad positive band, on the other hand, is expected to increase linearly with the percentage reconstitution. On the basis of these considerations a satisfactory explanation of the CD reconstitution experiments could be given. Since it appears to be impossible to explain the titration experiments without the quadratic term, we conclude that chromophore-chromophore interactions play an important role. No significant changes in secondary structure upon reconstitution could be detected consistent with our binding model which neglects cooperativity.

    Notes: Abstract We observed that particles, suspended in an electrolyte and brought into crossed magnetic and electric fields of low intensities, will deviate in the central part of the electrophoresis chamber of a standard Zeiss Cytopherometer with a component vertical to both fields. The direction and magnitude, however, were sharply at variance with what would be expected by the action of the Lorentz force (EMF) on the surface of the particles. The magnitude of the deviation depends upon the magnetic and electric field strength, the ion concentration of the suspension medium and the geometry of the chamber. The movement of the particles is due to streaming of the electrolyte which is mainly caused by inhomogeneities of the electric field in the electrophoresis chamber. The magnitude of the effect is high enough to occur under physiological conditions. Magneto-electrophoretic streaming might eventually act as a transducer mechanism which could explain the ability of some animals to orientate themselves in the geomagnetic field.

    Description / Table of Contents: Résumé Dans une premiere étape, on a procédé chez 4 chiens à la construction d'une poche de Heidenhain (PH) et chez 3 autres à une poche de Pavlov (PP). Chez 2 des chiens porteurs d'une poche de Pavlov on a aussi installé une canule oesophagienne. Chez tous les chiens, dans une 2ème et 3ème étape on a procédé respectivement à une vaguotomie proximale sélective (VPS) et à une pyloroplastie sous-muqueuse (P). Chez les chiens porteurs d'une PP et d'une canule oesophagienne, on a procédé à un test de repas simulé avant et après la VPS. Chez tous les autres animaux on a procédé à l'épreuve du repas après la completion des 3 étapes chirurgicales. Le repas simulé a produit une augmentation significative des taux de gastrine après la VPS mais la sécrétion d'acide a été abolie dans la PP. Après repas, les taux de gastrine se sont élevés significativement chez tous les chiens. Dans la PH, la sécrétion d'acide a été proportionnelle aux taux de gastrine, tandis dans la PP, après VPS, la sécrétion d'acide a été inhibée, malgré l'élévation post-brandiale des taux de gastrine. La construction d'une pyloroplastie a subséquemment réduit de faÇon significative la mise en circulation de gastrine chez tous les chiens. Cepandant, la sécrétion d'acide a diminué significativement seulement chez les chiens porteurs de PH.
    Notes: Abstract In a first operative stage, 4 dogs were prepared with Heidenhain pouches (HP) and 3 with Pavlov pouches (PP), 2 of the latter with an additional esophageal cannula. In a second stage all dogs had a selective proximal vagotomy (SPV), and in a third stage a submucosal pyloroplasty (P). Sham feeding experiments were carried out on the 2 dogs with PP and an esophageal fistula before and after SPV. Feeding tests were performed in all animals after the three operative procedures. Gastrin release after sham feeding increased significantly after SPV, whereas PP acid secretion was abolished. Gastrin release after feeding increased significantly after SPV in both HP and PP dogs. Acid secretion from HP was correlated with the changes in postprandial serum gastrin levels. PP acid secretion was diminished after SPV, in spite of the elevated gastrin levels. Subsequent pyloroplasty induced a significant decrease in the postprandial gastrin levels in both HP and PP dogs. However, acid secretion decreased significantly only in HP dogs.

    Description / Table of Contents: Zusammenfassung In vitro konnte gezeigt werden, daß inaktiviertes Maul- und Klauenseuchevirus (MKSV) bei einer NaCl-Konzentration von 0,1–0,15 mol nicht an DEAE-D/Kieselgur-Säulen adsorbiert wurde. Da derartige NaCl-Konzentrationen in DEAE-D/MKSV-Vakzinen und im Tierkörper vorhanden sind, dürfte die Adsorption von Virusantigen an DEAE-D nicht für dessen Adjuvanseffekt verantwortlich sein. Bei Schweinen ist an Hand des Auftretens von neutralisierenden Antikörpern und des Schutzes gegenüber Testinfektion nachgewiesen worden, daß DEAE-D seine optimale Adjuvanswirkung entfaltet, wenn es zusammen mit inaktiviertem MKSV als Vakzine injiziert wird. Daneben trat jedoch eine gute Adjuvanswirkung (75–100%ige Gruppenimmunität) bei etwa der Hälfte und eine mittlere Adjuvanswirkung (50–70%ige Gruppenimmunität) bei etwa einem Viertel der Impflinge auch dann auf, wenn DEAE-D örtlich und zeitlich vom inaktivierten Virus getrennt appliziert wurde. Dabei scheint es gleichgültig zu sein, ob DEAE-D oder inaktiviertes Virus zuerst gegeben wurde, jedoch war offensichtlich ein Zeitraum von 48 Stunden bzw. 4 Tagen zwischen beiden Injektionen günstiger als ein solcher von 24 Stunden. Hinsichtlich der Bildung von neutralisierenden Antikörpern ist die Situation mit jener bei der Immunität vergleichbar. Die einzige Abweichung besteht darin, daß ein Zeitraum von 24 Stunden zwischen der Applikation von DEAE-D und inaktiviertem Virus bzw. umgekehrt ebenso wirksam war als ein solcher von 48 Stunden bzw. 4 Tagen. Die Ergebnisse werden im Hinblick auf den möglichen Mechanismus des Adjuvanseffektes von DEAE-D auf zellulärer Ebene diskutiert.
    Notes: Summary In vitro it was shown that adsorption of inactivated FMDV onto DEAE-D kieselgur columns did not occur in the presence of 0.1–0.15m NaCl. These NaCl concentrations are present in DEAE-D/FMDV vaccines and in the tissues of animals. Therefore, adsorption of virus antigen does not appear to be responsible for the adjuvant effect of DEAE-D. In pigs it was demonstrated that DEAE-D exerts its optimal adjuvant effect, as measured by the formation of neutralizing antibodies and protection against challenge infection, when injected together with inactivated FMDV as vaccine. Apart from this, a good adjuvant effect (group immunity 75–100 per cent) was evoked in about one half and a moderate effect (group immunity 50–70 per cent) in about a quarter of the inoculated animals even if DEAE-D was separately injected locally and temporally from the inactivated virus. With regard to immunity it apparently does not matter whether DEAE-D or inactivated virus was given first, but an interval of 48 hours or 4 days between injection seemed to be more favourable than one of 24 hours. With regard to the formation of neutralizing antibodies the situation is comparable to that of immunity with the exception that a time interval of 24 hours between the application of DEAE-D and inactivated virus or vice versa was as good as that of 48 hours or 4 days. The results are discussed in regard to the possible mechanism of the adjuvant effect of DEAE-D on the cellular level.

    Description / Table of Contents: Summary The statistical relationship between the titers of neutralizing antibodies and the immunity of 706 pigs vaccinated against FMD was studied. This was done for each of four virus strains separately. Whereas no correlation between both test systems could be detected in case of the strains A5 Westerwald, C Detmold and O1Santander, a significant correlation was ascertained for the strain O1Kaufbeuren. Because of the different findings depending on the virus strain under study it was concluded that the antibody titer alone does not provide a useful measure for potency testing of FMD-vaccines for pigs.
    Notes: Zusammenfassung Die Titer der neutralisierenden Antikörper im Serum und der Immunstatus von 706 gegen die MKS vakzinierten Schweinen wurden, aufgeschlüsselt in die zur Vakzineproduktion verwendeten vier Virusstämme, statistisch in Beziehung gebracht. Dabei bestand bei den Virusstämmen A5W, CD und O1S keine Korrelation zwischen der Höhe der Neutralisationstiter und dem Prozentsatz an geschützten Tieren. Für den Stamm O1K wurde jedoch eine Korrelation zwischen der Höhe der Antikörpertiter und dem Prozentsatz der geschützten Tiere gefunden. Wegen der unterschiedlichen Befunde bei den einzelnen Virusstämmen wird die Bestimmung der Antikörpertiter allein zur Wirksamkeitsprüfung von MKS-Schweinevakzinen als nicht geeignet erachtet.

    Notes: Summary A thermolabile as well as thermosensitive mutant of vesicular stomatitis virus, VSV-tl-17, could be thermostabilized by phenotypic mixing with avian RNA tumor viruses (ATV). Biological, immunological and morphological studies revealed that this effect is due to a replacement of the thermolabile projections of the VSV by the avian tumor viral projections. The arrangement of projections of VSV and ATV on phenotypically mixed viria was studied by electron microscopy. A-type particles were detected in chick embryo cells which were doubly infected by ATV and VSV. Their intracytoplasmic appearance seemed to be the result of a disturbed maturation of ATV due to the relative deficiency of envelope proteins which were depleted by VSV maturation in the course of phenotypic mixing.
