    Description / Table of Contents: Zusammenfassung Die Synthese von [Deutero-(2-oxo-propyl)]-propylnitrosamin, (Deutero-methyl)-propylnitrosamin und den entsprechenden 3H-markierten Verbindungen wird beschrieben. Die in vivo Do-Applikation von [3H-(2-oxo-propyl)]-propylnitrosamin und von (3H-Methyl)-propylnitrosamin führte zur Bildung von (3H-7-Methyl)-guanin in den Nucleinsäuren der Rattenleber, was darauf hindeutet, daß diese Verbindungen natürliche Metaboliten des Di-n-propylnitrosamins sind. 2-Oxopropyl-propylnitrosamin läßt sich in wäßrig-alkalischem Medium zu Methyl-propylnitrosamin verseifen und zeigt damit eine gewisse Analogie zu β-Dicarbonylverbindungen.
    Notes: Summary The synthesis of [Deutero-(2-oxo-propyl)]-propylnitrosamine, (Deutero-methyl)propylnitrosamine and of the corresponding tritiated compounds is described. In vivo application of both [3H-(2-oxo-propyl)]-propylnitrosamine and (3H-methyl)-propylnitrosamine led to the formation of (3H-7-methyl)-guanine in nucleic acids of rat liver. This finding indicates that these compounds could be the metabolites of di-n-propylnitrosamine (Krüger, 1971). It could also be demonstrated that 2-oxo-propyl-propylnitrosamine can easily be hydrolysed to methylpropylnitrosamine under alkaline conditions and therefore resembles in some respects to a β-dicarbonyl compound.

    Description / Table of Contents: Zusammenfassung Die chronische Wirkung von 2-Oxo-propyl-propylnitrosamin (2-OPPN) wurde an Sprague-Dawley Ratten untersucht. Die Tiere zeigten vorwiegend hepatocelluläre Carcinome, die Wirkung dieses Carcinogens war dosisabhängig. Die Ergebnisse wurden unter der Annahme diskutiert, daß 2-OPPN möglicherweise ein Metabolit des Di-n-propylnitrosamins ist.
    Notes: Summary The chronic effect of 2-oxo-propyl-propylnitrosamine (2-OPPN) was examined in Sprague-Dawley rats. The animals showed mainly hepatocellular carcinomata; the effect of the carcinogen was dose dependent. The findings are discussed under the assumption that 2-OPPN might be a metabolite of di-n-propylnitrosamine.
