Summary The cytostatic compounds Bis- (β-Chloroethyl)-methylamineoxide-hydrochloride (N-Oxide-Mustard), 2,5-Bis-ethyleneiminobenzoquinone-1,4 (BBQ) and 4,4′-Dioxy-α,β-diethylstilbene (DDS) have been investigated with regard to their induction of recessive sex-linked lethals in Drosophila melanogaster. Furthermore the mutagenic action of N-Oxide-Mustard on several stages of immature spermatozoa has been determined. N-Oxide-Mustard and BBQ induce a high rate of mutations being 16 respectively 4 times as high as the percentage of spontaneous mutations. DDS, however, has no mutagenic qualities. Shortly after meiosis the immature spermatozoa are most sensitive to the mutagenic action of N-Oxide-Mustard. The mechanism of cytostatic and mutagenic action of the various compounds is discussed and compared with cellular reactions following X-irradiation. The danger of a damage of the hereditary material caused by the clinical application of cytostatic compounds is emphasized.
