    Notes: The effect of nicotinamide on unscheduled DNA synthesis was studied in resting human lymphocytes. In cells treated with UV irradiation or with MNNG, nicotinamide caused a two-fold stimulation of unscheduled DNA synthesis and retarded the rate of NAD+ lowering caused by these treatments. Nicotinamide also reduced the burst of poly(ADP-ribose) synthesis caused by MNNG treat-ment. Thus under conditions that it enhances unscheduled DNA synthesis, nicotinamide causes marked effects on the metabolism of NAD+ and poly(ADP-ribose). The effect of nicotinamide on unscheduled DNA synthesis was shown to be independent of protein or polyamine synthesis.

    Notes: The 13C chemical shifts of 2-substituted and 2,6-disubstituted anthraquinones have been determined and assigned. The C-1, 2, 3, 4, 13 and C-14 chemical shifts of 2-substituted anthraquinones are correlated with the chemical shifts of monosubstituted benzenes. A three-parameter correlation with Swain and Lupton's F and R parameters and Schaefer's Q parameter provides relationships for the prediction of all chemical shifts of 2-substituted anthraquinones from the substituent parameters. Q values for the SCH3, OCOCH3, C2H5 and C(CH3)3 groups are proposed. The two types of correlations are compared for predicting chemical shifts.

    Notes: The spectra of various 1-substituted and 1,5-disubstituted anthraquinones have been studied and the chemical shifts of the different carbon nuclei determined. The chemical shifts previously reported for 1-Me-anthraquinone have been corrected and reassigned. The C-1, C-4 and C-13 shifts of 1-substituted derivatives are correlated with the chemical shifts of monosubstituted benzenes. Deviations from the regression lines can be explained by the existence of steric factors and hydrogen bonding. A three-parameter correlation with Swain and Lupton's F and R and Schaefer's Q provides relationships for the prediction of all chemical shifts of 1-substituted anthraquinones when the substituents have a cylindrical symmetry.

    Notes: Drosophila Kc cells exposed to physiological doses of the moulting hormone, β-ecdysone, elongate, become motile, and subsequently aggregate. This pattern of morphogenesis was found to require the assembly of a microtubular cytoskeleton. Tubulin content was significantly increased in hormone-treated cells when compared to controls, as measured by a 3H-colchicine-binding assay. However, determinations of rates of tubulin synthesis and breakdown revealed no difference between control and hormone-treated cells for either parameter. When tubulin content was assayed by methods that do not depend on colchicine-binding activity, no difference between hormone-treated and control cells was observed. These results are discussed in terms of a model in which β-ecdysone affects the distribution of tubulin in “assembly-active” and “assembly-inactive” pools.

Die Ergebnisse bestätigen die wesentlichsten Eigenschaften des “blob” -Bildes, d. h. den Einfluß der nicht-uniformen Ausdehnung der Kette auf die Relaxationszeiten und die Intrinsic-Viskosität, unterscheiden sich im Detail jedoch von den Ergebnissen von DAUD und JANNINK sowie WEILL und DES CLOIZEAUX. Entgegen den Voraussagen wird ein zweites Plateau in [η(ω)] nicht beobachtet.
    Notes: The dynamics of polymer solutions are consistently treated by the BIXON-ZWANZIG theory including both hydrodynamic and excluded volume interactions. The results confirm generally the essential features of the “blob” picture in polymer dynamics, i.e. the influence of non-uniform chain expansion on the relaxation time spectra and the intrinsic viscosity, but differs in detail from results by DAOUD and JANNINK and WEILL and DES CLOIZEAUX. Especially no second plateau in [η(ω)] is observed.

    Description / Table of Contents: Suspensions of polyacrylonitrile in acrylonitrile were prepared by polymerization in bulk, and the morphology of suspended particles was investigated. In stirred systems the suspensions exhibited large differences in rheological properties under comparable degrees of conversions on varying the experimental conditions. The variation of the properties is caused by the formation of two kinds of polyacrylonitrile particles which differ in shape, size, and density. The structures of the particles, however, are identical.
    Notes: Durch Massepolymerisation wurden Suspensionen von Polyacrylnitril in Acrylnitril hergestellt und die Morphologie der suspendierten Teilchen untersucht. In gerührten Systemen Wiesen diese Suspensionen bei Variation der Versuchsbedingungen große Unterschiede der rheologischen Eigenschaften bei vergleichbaren Umsätzen auf. Die unterschied lichen Eigenschaften beruhen auf der Bildung von zwei Typen der Polymerteilchen, die sich in Form, Größe und Dichte bedeutend unterscheiden. Der Aufbau der Teilchen ist jedoch gleich.

    Notes: Potentiometric titration curves of oxidized and reduced horse heart cytochrome c in 0.15M KCl at 20°C have been obtained by timed titration (0.125-0.500 μmol/sec) from the isoionic points (pH 10.2-10.4) to pH 3 and back to the isoionic point. Computer-assisted (PROPHET) data acquisition and blank corrections give curves with good precision with a maximum standard deviation of 0.3 groups for an average error of 1%. The potentiometric titration curve of reduced cytochrome c is reversible within the precision of the method and for the pH range studied. The potentiometric curves for oxidized cytochrome c titrated upscale (pH 3-10) and downscale (pH 10-3) are not reversible. However, they show the same ionization behavior after the initial downscale titration. This is probably the result of a conformational change. Comparison of the data herein reported with the titration curves of oxidized cytochrome c already published by others indicates good agreement on the basis of a normalization of the concentration of protein or on the basis of 25 titrable groups between the acid end point and the isoionic pH.Titration of the 2 μmol imidazole in the upscale or downscale direction gives the correct analytical concentration and pK′ after correction for the solvent titration. Titration of reduced cytochrome c in the presence and absence of an additional equivalent of imidazole gave a difference titration curve, which indicates that a group on the protein shifts from pK′ 5.8 to pK′ 5.3 in the presence of imidazole. The pK′ of imidazole, in the presence of the protein, remains at a nearly normal value of 7.34.

    Notes: The endogenous lectins discoidins I and II are believed to be primary components of the morphogenetic cell cohesion system of D discoideum. We have developed two immunochemical methods to analyze the association of the discoidins with the cell surface. One method is a two-stage specific antibody binding assay in which intact cells are incubated on ice with rabbit serum (either control serum or antidiscoidin I and II), washed, then incubated with 125I-Protein A. Specific antibody binding is defined as the difference between percent radioactivity bound with antidiscoidin versus control serum during the first stage. Substantial specific binding was observed with developed A3 cells but not with vegetative cells, and nearly all of the activity could be removed by pread-sorption of the antiserum with discoidin-Sepharose. As a complementary method, quantitative immunoadsorption analysis was performed in which we tested the ability of intact cells to remove antibodies reactive with purified 125I-discoidin I or II. Developed cells, but not vegetative cells, were capable of adsorbing antibodies reactive with discoidin I as well as those reactive with discoidin II. This represents the first demonstration that both lectins are present on the surface of cohesive cells.These procedures, coupled with other methods to analyze soluble discoidin in cell extracts, were used to study discoidin expression in wild type cells and in two newly isolated aggregation-defective mutants. Strain EB-32 fails to aggregate and displays little or no discoidin in cell extracts or at the cell surface. On the other hand, strain EB-18 forms loose amorphous mounds, and expresses substantial quantities of the discoidins, both in cell extracts and at the cell surface. These mutants should prove valuable in studying the organization and regulation of discoidins I and II at the surface of aggregating cells.
