    Description / Table of Contents: Summary Protein A from Staphylococcus aureus, a cell wall protein with high affinity binding properties for IgG-type antibodies, has been labeled with peroxidase to form a stable immunohistological tracer molecule of relatively low molecular weight. It has been used for demonstration and titration of antinuclear antibodies in SLE sera on mouse liver sections in an indirect technique. The findings were consistent with those obtained by immunofluorescence and by staining with peroxidase-coupled anti-IgG. In contrast to immunofluorescence, the stained sections could be mounted and stored for documentation. In comparison, unspecific tissue adsorption and staining could be minimized by addition of glucose, galactose, and mannose as well as bovine serum albumine to the buffer containing Protein A-peroxidase.
    Notes: Zusammenfassung Protein A von Staphylococcus aureus, ein Zellwandprotein mit hoher Affinität für Antikörper vom Type IgG, wurde mit Peroxidase markiert. Es entstand ein stabiles Tracer-Molekül von relativ niedrigem Molekulargewicht für den immunohistologischen Nachweis von gewebsgebundenen Antikörpern. In einer indirekten Technik wurden auf Mäuselebergewebe antinukleäre Antikörper aus Seren von Patienten mit Systemischem Lupus Erythematodes nachgewiesen und titriert. Die Befunde stimmten mit denjenigen bei Immunfluoreszenz und bei Immunoenzymtechnik mit Peroxidase-markiertem anti-IgG überein. Im Gegensatz zur Immunfluoreszenz können die Präparate nach der Immunoenzymreaktion zur Dokumentation aufbewahrt werden. Die bei allen Techniken auftretende unspezifische Adsorption von Tracermolekülen im Gewebe konnte durch Zusatz von Glucose, Galactose und Mannose zur Protein A-Peroxidase-Suspension deutlich vermindert werden.

    Description / Table of Contents: Zusammenfassung Überprüft man in Histiocytomen mit Hilfe des Nachweises der alkalischen Phosphatase den Verlauf der Capillaren, so überrascht die große Zahl der Gefäße. Von einem Zentrum erstrecken sich diese durch das ganze Histiocytom, selbst die über dem Histiocytom unter der Epidermis gelegenen Capillaren sind gewuchert. Histochemische und elektronenmikroskopische Untersuchungen zeigen aber, daß die Mehrzahl der Tumorzellen keine Gefäßzellen sind, sondern Fibroblasten, Histiocyten und Mastzellen. Die jüngeren Fibroblasten gleichen Elementen, die man auch bei der Wundheilung findet und die man als C-Fibroblasten bezeichnet hat.
    Notes: Summary In histiocytomas the alkaline phosphatase technique reveals a surprising number of capillaries. The capillaries branch out from the center to the periphery of the histiocytoma and even the capillaries beneath the epidermis proliferate. The cellular elements outside the capillaries, however, are fibroblasts, histiocytes and mast cells, as is revealed by various histochemical and electron microscopical examinations. Histiocytomas of short duration differ from older histiocytomas in that the former contain fibroblasts similar to those found in healing wounds (C-fibroblasts).

    Notes: Summary Cytochrome P-450-dependent arylhydrocarbon-hydroxylase (AHH) activity and inducibility by benzanthracene (BA) was measured in cultured guinea pig and human epidermal cells. Basal AHH-activity (AHHb) in guinea pig epidermal cells was much higher than in human epidermal cells. AHHb in guinea pig epidermal cells was directly related to the labeling index and decreased to the original level between the 5th and 7th day of cell culturing. On the other hand, the induction-ratio of AHH reached its maximum level when the number of cells began to rise (proliferation phase) and remained high at day 7 of the cell culture. These results suggest a cell growth dependent activity and inducibility of carcinogen-metabolizing enzymes, such as AHH, in isolated epidermal cells.
