    Notes: A systematic survey of seven parallel α/β barrel protein domains, based on exhaustive structural comparisons, reveals that a sizable proportion of the αβ loops in these proteins - 20 out of a total of 49 - belong to either one of two loop types previously described by Thornton and co-workers. Six loops are of the αβ1 type, with one residue between the α-helix and β-strand, and 13 are of the αβ3 type, with three residues between the helix and the strand. Protein fragments embedding the identified loops, and termed αβ connections since they contain parts of the flanking helix and strand, have been analyzed in detail revealing that each type of connection has a distinct set of conserved structural features. The orientation of the β-strand relative to the helix and loop portions is different owing to a very localized difference in backbone conformation. In αβ1 connections, the chain enters the β-strand via a residue adopting an extended conformation, while in αβ3 it does so via a residue in a near α-helical conformation. Other conserved structural features include distinct patterns of side chain orientation relative to the β-sheet surface and of main chain H-bonds in the loop and the β-strand moieties. Significant differences also occur in packing interactions of conserved hydrophobic residues situated in the last turn of the helix. Yet the α-helix surface of both types of connections adopts similar orientations relative to the barrel sheet surface. Our results suggest furthermore that conserved hydrophobic residues along the sequence of the connections, may be correlated more with specific patterns of interactions made with neighboring helices and sheet strands than with helix/strand packing within the connection itself. A number of intriguing observations are also made on the distribution of the identified αβ1 and αβ3 loops within the α/β-barrel motifs. They often occur adjacent to each other; αβ3 loops invariably involve even numbered β-strands, while αβ1 loops involve preferentially odd β-strands; all the analyzed proteins contain at least one αβ3 loop in the first half of the eightfold α/β barrel.     Notes: The X-ray structure of Escherichia coli TEM1β-lactamase has been refined to a crystallorgphic R-factor of 16.4% for 22,510 reflections between 5.0 and 1.8 Å resolution; 199 water molecules and 1 sulphate ion were included in refinement. Except for the tips of a few solvent-exposed side chains, all protein atoms have clear electron density and refined to an average atomic temperature factor of 11 Å2. The estimated coordinates error is 0.17 Å. The substrate binding site is located at the interface of the two domains of the protein and contains 4 water molecules and the sulphate anion. One of these solvent molecules is found at hydrogen bond distance from S70 and E166. S70 and S130 are hydrogen bonded to K73 and K234, respectively. It was found that the E. coli TEM1 and Staphylococcus aureus PC1 β-lactamases crystal structures differ in the relative orientations of the two domains composing the enzymes, which result in a narrowed substrate binding cavity in the TEM1 enzyme. Local but significant differences in the vicinity of this site may explain the occurrence of TEM1 natural mutants with extended substrate specificities. © 1993 Wiley-Liss, Inc.

    Notes: The deficiency of the lysosomal protective protein/carboxypeptidase L (CARB L) causes the lysosomal storage disorder, galactosialidosis, characterized by neuraminidase and β-galactosidase deficiencies in patients' cells. The three enzymes form a complex inside the lysosome, and the neuraminidase and β-galactosidase deficiencies are secondary to CARB L deficiency. Sequence similarity and common enzymological properties suggest that the protomeric tertiary structure of CARB L is conserved within a family of serine carboxypeptidases which includes the yeast carboxypeptidase Y, killer expression I gene product and several plant carboxypeptidases. We used this homology to build a model of the CARB L structure based on the recently published X-ray atomic coordinates of the wheat carboxypeptidase II (CPDW-II) which shares 32% primary structure identity with CARB L. Small insertions and deletions were accommodated into the model structure by energy minimization using the DREIDING II force field. The Cα atomic-coordinates of the final CARB L model have a RMS shift of 1.01 Å compared to the corresponding conserved residues in the CPDW-II template structure. The correct orientation of the homologous catalytic triad residues Ser150, His429 and Asp392, the potential energy calculations and the distribution of hydrophobic and hydrophillic residues in the structure all support the validity of the CARB L model. Most missense mutations identified in galactosialidosis patients were located in secondary structural elements except for the Tyr211→Asn mutation which is in a loop. The other mutant residues have their side chains deeply buried in the central β-sheet of the model structure except for the Phe412→Val mutation which is located in the dimer interface. The predicted effects of specific mutations on CARB L structural stability correlates well with recently published transient expression studies of mutant CARB L (Shimmoto, M. et al., J. Clin. Invest., 91:2393-2399, 1993). © 1994 John Wiley & Sons, Inc.

    Notes: Nucleophilic substitution of PVC with sodium thiophenate was carried out in cyclohexanone solution at 5, 25, 40, 60, and 70°C. The initial rate obeys an Arrhenius law from 25 to 60°C, with an activation energy of 70 kJ/mol. Conversion limits are observed which strongly depend on the temperature. The stereoselectivity of the reaction with respect to the configurational triads does not depend on the temperature: the distribution of configurations is only dependent on the conversion. Assuming an SN2 substitution mechanism governed by steric factors, the Monte Carlo simulation procedure described in a prior study is shown to give a good account for all temperatures above 40°C assuming for the mm, mr or rm, and rr triads a reactivity such as Rmm = 2 Rmr and Rrr nil at low temperature and very low at temperatures ≥ 40°C. The low conversion limits observed at 5 and 25°C cannot be explained by a limited accessibility of a part of the polymer. Finally, it is shown that the elimination reaction, which remains limited, does not interfere with the substitution process.

    Notes: Polymers can be ozonized with an ozone/oxygen mixture to create peroxides and hydroperoxides on the polymer chain. These heat-sensitive functional groups can be used to initiate radical polymerization of vinyl monomers and give graft copolymers. The properties of these copolymers are well described in literature; however, we notice that available data concerning decomposition rate constants are inconsistent. Decomposition rate measurements, time of assay of free radicals and stability of the ozonized polymers do not give the same results. In this work, we studied the kinetics of the styrene polymerization initiated by an ozonized ethylene vinyl acetate copolymer. Consequently, the f·kd product can be calculated (f is the efficiency and kd the decomposition rate constant). The ozonized copolymer is not similar to usual radical initiators. The initial viscosity of the solution has an important effect on the calculation of the kinetic values. Activation energy and Arrhenius coefficient are different than the one obtained for the usual initiators. © 1994 John Wiley & Sons, Inc.

    Notes: Concerning the study on the relation between structural parameters and reactivity in ring-opening metathesis polymerization of cyclo-olefins, the “living” polymerization of norbornene initiated by Schrock's-type complexes was considered as a reference and studied from the kinetic point of view. First kinetic orders with respect to both monomer and active species allow the values of absolute rate constants of propagation to be determined. The thermodynamic parameters obtained from kinetic experiments performed at different temperatures seem to indicate that monomer coordination and metallacycle formation are rate-determining steps in the process studied. © 1994 John Wiley & Sons, Inc.

    Notes: The feasibility of using simulated moving bed technology (SMB) for chiral separation on cellulose triacetate is demonstrated on the preparative scale: 1 kg of a chiral epoxide has been separated. On comparing SMB technology with conventional liquid chromatography it turns out that the main advantage of SMB lies in the significant reduction of mobile phase consumption. The process design for SMB is made theoretically and the predictions are confirmed by our pilot study. © 1993 Wiley-Liss, Inc.

    Notes: A selective antibody to (S)-propranolol enantiomer was produced in rabbits by immunization with a new conjugate of N-aminopropylpropranolol-albumin. A hapten was first prepared by condensing (S)-propranolol or the racemate with 3-bromopropylphthalimide followed by hydrazinolysis, and the resulting compound conjugated to serum albumin by means of a glutaraldehyde- or carbodiimide-mediated reaction. Rabbits were immunized, and titres and specificity of antibodies were determined by ELISA. The antibodies obtained were tested with (S)-, (R)-, (R, S)-propranolol, and other structural analogs. Selective (S)-antibodies recognized (S)-propranolol 20 times more avidly than (R)-isomer while an antiserum against (R, S)-propranolol recognized both (S)- and (R)-isomers to about the same degree. ©1993 Wiley-Liss, Inc.

    Notes: The optimization of a chemical synthesis aims at an adjustment of the operating conditions, i.e. pH, temperature and initial concentrations, in order to obtain maximum yield, minimum operating costs or similar. The optimization effort may be considerable so that, in many cases, its cost cannot be justified, especially for small volume products. This paper introduces a new strategy, namely to establish the optimum operating conditions with a relatively small effort. The new approach is illustrated by the synthesis of 2-furyl oxirane. In the first step, a simplified mechanistic process model has to be determined on the basis of easily accessible experimental data. Initial and final concentrations of all reacting components, reaction time and temperature must be known. A two-step evaluation yields the stoichiometric coefficients of all the occurring reactions as well as the parameters of the Arrhenius model. This so-called trend model is used in the optimization of the operating conditions. It is not only important to know the optimum operating conditions but also to maintain them as close as possible in practical operation. An adaptive controller is well suited to such an application and its use is discussed.
