    Abstract: As serious side effects affected recent virus-mediated gene transfer studies, novel vectors with improved safety profiles are urgently needed. In the present study, replication-deficient retroviral vectors based on feline foamy virus (FFV) were constructed and analyzed. The novel FFV vectors are devoid of almost the complete env gene plus the internal promoter - accessory bel gene cassette including the gene for the viral transcriptional transactivator Bel1/Tas. In these Bel1/Tas-independent vectors, expression of the lacZ (beta-galactosidase) marker gene is directed by the heterologous, constitutively active human ubiquitin C promoter (ubl). Env-transcomplemented vectors have unconcentrated titers of more than 10(5) transducing units/ml. The vectors allow efficient transduction of a broad array of diverse target cells, which can be increased by repeated vector exposure. However, the number of lacZ marker gene expressing cells decreased slightly upon serial passages of the transduced cells. Vectors carrying a self-inactivating (SIN) deletion of the TATA box and most parts of the viral promoter were not rescued by wt FFV whereas those with the intact or a partially deleted promoter were readily reactivated. This finding indicates that the viral promoters are in fact non-functional, pointing to a highly advantageous safety profile of these new FFV-ubi-lacZ-SIN vectors

    Abstract: Rodent autonomous parvoviruses (PVs) are endowed with oncotropic properties and represent virotherapeutics with inherent oncolytic features. This work aimed to evaluate the capacity of Minute Virus of Mice (MVMp) to act as an adjuvant stimulating a mouse glioblastoma-specific immune response. MVMp was shown to induce cell death through apoptosis in glioma GL261 cells. Antigen-presenting cells (APCs) provide the initial cue for innate and adaptive immune responses, and thus MVMp-infected GL261 cells were tested for their ability to activate dendritic cells (DCs) and microglia (MG), two distinct cell types that are able to act as APCs. MG and discrete DC subsets were activated after co-culture with MVMp-infected glioma GL261 cells, as evidenced by upregulation of specific activation markers (CD80, CD86) and release of proinflammatory cytokines (tumor necrosis factor-alpha and interleukin-6). The in vivo analysis of immunodeficient and immunocompetent mice revealed a clear difference in their susceptibility to MVMp-mediated tumor suppression. Immunocompetent mice were fully protected from tumor outgrowth of GL261 cells infected ex vivo with MVMp. In contrast, immunodeficient animals were less competent for MVMp-dependent tumor inhibition, with only 20% of the recipients being protected, arguing for an additional immune component to allow full tumor suppression. In keeping with this conclusion, immunocompetent mice engrafted with MVMp-infected glioma cells developed a level of anti-tumor immunity with isolated splenocytes producing elevated levels of interferon-gamma. In rechallenge experiments using uninfected GL261 cells, we could show complete protection against the tumor, arguing for the induction of a T-cell-mediated, tumor-specific, long-term memory response. These findings indicate that the anticancer effect of PVs can be traced back not only for their direct oncolytic effect, but also to their ability to break tumor tolerance.

    Description: Due to the increasing number of long-term cancer survivors there is a growing necessity of facilitating occupational reintegration in the context of cancer rehabilitation. We report evaluation results of a specifically developed occupation oriented rehabilitation concept within inpatient medical cancer rehabilitation. Even less burdened patients reported support and counselling needs. On the other hand, rehabilitation participants who did not return to work within 12 months after rehabilitation initially reported high distress levels. For cancer rehabilitation are therefore specific offers such as occupation-related assessments, stress management or coaching recommendable for participants with lower distress levels, while the subgroup of highly distressed patients requires intensive training offers. Participants of the intervention group of this study tended to return to work earlier and assessed their occupational performance and satisfaction at follow up more positive than the comparison group.
    Description: Auf Grund der zunehmenden Zahl von Langzeitüberlebenden einer Krebserkrankung wächst die Notwendigkeit berufliche Reintegration im Rahmen der onkologischen Rehabilitation zu erleichtern. Berichtet werden Evaluationsergebnisse zu einem spezifisch entwickelten medizinisch beruflich orientierten Rehabilitationskonzept in der stationären onkologischen Rehabilitation.Auch wenig belastete Patienten gaben subjektiven Unterstützungs- und Beratungsbedarf an. Umgekehrt zeigt sich, dass Rehabilitandinnen und Rehabilitanden, die 12 Monate nach der Rehabilitation nicht zur Arbeit zurückgekehrt sind, bereits initial hoch belastet sind. Für die onkologische Reha empfehlen sich deshalb spezifische Angebote für wenig belastete Rehabilitanden, wie berufliche Standortbestimmung, Stressbewältigung oder Coaching, während die Subgruppe der hochbelasteten intensive Trainings- und nachhaltige Angebote benötigt. Patienten der Interventionsgruppe kehrten in dieser Studie, tendenziell früher an den Arbeitsplatz zurück und schätzten zum Katamnesezeitpunkt ihre subjektive berufliche Leistungsfähigkeit und Zufriedenheit nachhaltig höher ein als die Vergleichsgruppe.

    Notes: Abstract Certain small networks of spiking interneurons are shown to produce strong fluctuations when modeled with neuromimes. The reasons for these fluctuations are demonstrated and discussed. When one or more of the neuromimes in the circuit were replaced by non-spiking neuromimes, the fluctuations were strongly reduced or disappeared completely. We assume that one of the important functions of nonspiking neurons is to provide precise analog computation while avoiding fluctuation.

    Notes: Abstract The improved prognosis and survival statistics of both renal transplantation and dialysis have focused attention on the quality of life offered by these treatments. Using a standardized questionnaire, we assessed the quality of life of 612 patients undergoing renal replacement therapy at our center. Of these patients, 359 had been transplanted and 253 patients were on dialysis. Concerning the sociodemographic data, only the time on specific treatment was longer in dialysis patients than in transplanted patients (49.2 versus 55.6 months, P 〈 0.05). Most complaints were more common in dialysis patients than in transplanted patients. Only the side effects of medication were seen more in transplanted patients (P 〈 0.005). Life satisfaction was higher in transplanted patients than in dialysis patients. Dialysis patients were more anxious (P 〈 0.05) and more depressed (P 〈 0.001) than transplanted patients. Transplanted patients also felt that they had more social support than did dialysis patients. Overall life quality was almost equal between patients on hemodialysis and patients on peritoneal dialysis, and between patients on the waiting list for transplantation and those not on the waiting list. Despite a significantly better quality of life after renal transplantation, the percentage of patients working remained unchanged. (57.5 % versus 57.8 %, P = n.s.). We conclude that despite an improved quality of life after renal transplantation, these patients are economically not more productive than patients on dialysis.
