Abstract: This book is dedicated to the great scientist and outstanding individual Nikolay Wladimirovich Timofeeff-Ressovsky. The book brings together a number of brief stories/essays about Timofeeff-Ressovsky including “Stories told by himself”, and scientific chapters addressing his major research areas: genetics, radiobiology, radiation ecology and epidemiology, and evolution. Timofeeff-Ressovsky contributed to several fields of biology and established new directions of scientific research. He often repeated the phrase, which would later become famous: “Science should not be approached with the ferocity of wild animals”. In keeping with that philosophy, the issues discussed here are still open. Each scientific part starts with a current review; the chapters present leading scientific schools and views. The main theme discussed in the genetics part is mutation variability in the context of linear (replication, transcription, translation) and conformational template processes, and its dependence on phylogenetic group. In turn, the radiobiology chapters focus on the reorganization of DNA, cell, and population variability under low-dose irradiation, sparking indirect processes and adaptive response. The radiation ecology and epidemiology parts present data on the consequences of nuclear plants and related accidents for ecological systems and human beings. Here some approaches to estimating radiation risks are also offered. Evolution laws are demonstrated in the genomic universe, plant-microbe symbiosis, stabilizing and destabilizing (directional) selection. The last essay demonstrates the principles of organization operating in local animal populations, which are approached as social organisms of complex systemic nature. The chapter 'Radiation-Induced Aging and Genetic Instability of Mesenchymal Stem Cells: An Issue for Late Health Effects?' is available open access under a CC BY 4.0 license

Drug Resistance in Bacteria, Fungi, Malaria, and Cancer (2017)
Cham : Springer International Publishing
Keywords: Medicine ; Cancer Research ; Drug Resistance ; Cell Biology ; Bacteriology ; Biomedicine ; Drug Resistance ; Cell Biology ; Cancer Research ; Bacteriology ; Springer eBooks
Abstract: This book is a compilation of past and recent knowledge in the field of emerging drug resistance. The book covers major aspects of drug resistance in bacteria, fungi, malaria, and cancer. Human survival on earth is constantly threatened by disease and syndrome. From the early days, the aim of research in medicine was to find therapeutic agents that can improve the quality of human life. Although humans are dependent on natural compounds from early days their dependence of drugs increased excessively in last century. The advances in chemistry and biology have helped researchers to identify the drugs that have improved treatment of many diseases. The primary factor for treatment of these diseases is dependent on the efficacy of drugs available. The development of resistance to these drugs is one of the major hindrances. Although there are number of books available on this topic, “drug resistance” biology across kingdoms has never been discussed in a coherent way

Abstract: In the last decade, immunotherapy has revolutionized the practice of medicine in the field of oncology. This book, Immunotherapy, highlights the immunotherapeutic approaches currently in use and strategies to overcome the associated challenges. As comprehensive knowledge of basic immunology in the context of tumor biology is required to move further along the line of development in translational science, this book provides an overview of the cellular components of the immune system that interact with each other to provide a successful immune response. Immunotherapy offers promising antitumor activity across multiple cancer types as it does not attack the tumor but primarily revitalizes a suppressed immune system. It is beyond the scope of this book to discuss the immunotherapeutic approaches across the tumor board. However, the book features the role of immune-oncology in acute myeloid leukemia, non-small cell lung cancer, and sarcomas to showcase the recent advances in the management of cancer broadly classified as hematological malignancies, solid tumors, and tumors arising in the connective tissue. The book also discusses alternative formulations and combinatorial approaches to enhance the therapeutic index of these agents. As immunotherapeutic agents differ significantly from chemotherapeutic agents in response patterns and toxicity profiles, evaluating their safety and efficacy in clinical trials remains a challenge. In this book, we discuss the use of a variety of traditional and new immunotherapy criteria to evaluate response, their inadequacies, and clinical implications. The book also outlines how recent advances in medical imaging technology have the potential to provide high-dimensional data to improve decision support and individualize treatment selection and monitoring. Importantly, the book attempts to increase the awareness to immune related adverse events and the need for aggressive and judicious management of toxicities to improve treatment outcomes in patients on immunotherapy In short, this book provides a broad understanding of immunotherapy and ways to leverage the immune system to make inroads in the fight against cancer

Molecular Oncology: Underlying Mechanisms and Translational Advancements (2017)
Cham : Springer International Publishing

Description / Table of Contents: - Microtubule targeting agents in cancer therapy: Elucidating the underlying molecular mechanisms -- Adenosine signaling pathways as potential therapeutic targets in cancer disease -- Anticancer activities of Hippophae rhamnoides: Journey from Benchtop to the Bedside -- Emerging role of ncRNAs in cancer biology: techniques for diagnostic monitoring and potential ncRNA-based therapies -- Clinical and translational research on the pathophysiology of several ocular tumors types -- Exosomes and cancer: from diagnosis to therapy -- Blood-based Cancer Detection: Towards Solving Diagnostic Puzzle -- Non-coding RNAs, Critical Therapy Targets against Cancer - Detection of Virus-caused Cancer in Publicly Available Next Generation Sequencing Data -- TRAIL mediated signaling in prostate cancer -- Index
Abstract: Cancer is a multifaceted and genomically complex disease and data obtained through high throughput technologies has provided near complete resolution of the landscape of how genomic, genetic and epigenetic mutations in cancerous cells effectively influence homeostasis of signaling networks within these cells, between cancerous cells, tumor microenvironment and at the organ level. Increasingly sophisticated information has helped us in developing a better understanding of the underlying mechanisms of cancer, and it is now known that intra-tumor genetic heterogeneity, cellular plasticity, dysregulation of spatio-temporally controlled signaling cascades, and loss of apoptosis are contributory in cancer development, progression and the development of resistance against different therapeutics. It is becoming progressively more understandable that earlier detection of pre-existing or emerging resistance against different therapeutics may prove to be helpful in personalizing the use of targeted cancer therapy. Despite the fact that there is a continuously increasing list of books, being guest edited by researchers, books on the subject are often composed of invited reviews without proper sequence and continuity and designed for a particular readership. This book progressively shifts and guides the readers from basic underlying mechanisms to translational approaches to treat cancer

Abstract: This edited volume is about novel targeted therapies for the treatment of advanced neoplasms of the head and neck (HNSCC) with particular reference to oral squamous cell carcinomas and covers specific fields of cancer genetics and genomics for personalized medicine. The landmark studies from the HNSCC Cancer Genome Anatomy (TCGA) and the cancer Genome Atlas 2015 have revealed the multiplicity and diversity of genetic alterations in HNSCC. This remarkable body of information has elucidated novel druggable targets (eg. EGFR, mTOR, p53, survivin and aberrant microRNA) for therapeutic interventions in head and neck cancer. Readers will discover the importance of the biology and pathology of squamous cell carcinomas, the role of cancer stem cells and other factors that contribute to resistance to curative therapies such as hypoxia. We present an in-depth analysis of the HNSCC oncogenome describing the distinct molecular alterations in squamous cell carcinomas, which participate in specific molecular pathways whose dysregulation may contribute to most HNSCC cases. Final chapters of the book elucidate how these important cancer-driving molecular events and molecular pathways could be targeted to treat head and neck cancers and their therapeutic applications for targeted therapies based on clinical trials reported worldwide. This book is aimed at a broad audience, including scientists, physicians, surgeons, and post graduate students engaged in oncology and provide the readers with an up-to-date, interdisciplinary knowledge and tools to better understand the evolving areas of targeted gene therapy and the key concept of personalized medicine in head and neck cancer

Abstract: This volume provides the reader with an overview of the diverse functions of the RUNX family of genes. As highlighted in the introduction and several of the 29 chapters, humans and other mammals have three RUNX genes that are known to play specific roles in blood, bone and neuronal development. However, their evolutionary history has recently been traced back to unicellular organisms and their involvement in many well-known signaling pathways (Wnt, TGFb, Notch, Hippo) is indicative of a more general function in cell biology. Their documented roles in cell fate decisions include control of proliferation, differentiation, survival, senescence and autophagy. The pleiotropic effects of RUNX in development are mirrored in cancer, where RUNX genes can function as oncogenes that collaborate strongly with Myc family oncogenes or as tumour suppressor genes. In the latter role, they display hallmarks of both ‘gatekeepers’ that modulate p53 responses and ‘caretakers’ that protect the genome from DNA damage. Several chapters focus on the importance of these genes in leukemia research, where RUNX1 and CBFB are frequently affected by chromosomal translocations that generate fusion oncoproteins, while recent studies suggest wider roles for RUNX modulation in solid cancers. Moreover, RUNX genes are intimately involved in the development and regulation of the immune system, while emerging evidence suggests a role in innate immunity to infectious agents, including HIV. At the biochemical level, the RUNX family can serve as activators or repressors of transcription and as stable mediators of epigenetic memory through mitosis. Not surprisingly, RUNX activity is controlled at multiple levels, this includes miRNAs and a plethora of post-translational modifications. Several chapters highlight the interplay between the three mammalian RUNX genes, where cross-talk and partial functional redundancies are evident. Finally, structural analysis of the RUNX/CBFB interaction has led to the development of small molecule inhibitors that provide exciting new tools to decipher the roles of RUNX in development and as targets for therapy. This volume provides a compendium and reference source that will be of broad interest to cancer researchers, developmental biologists and immunologists

Human Chromosome Variation: Heteromorphism, Polymorphism and Pathogenesis (2017)
Singapore : Springer
Keywords: Medicine ; Medical genetics ; General practice (Medicine) ; Biomedicine ; Gene Function ; General Practice / Family Medicine ; Springer eBooks
Description / Table of Contents: Introduction -- Chromosome Heteromorphism -- Frequencies of Heteromorphisms -- Clinical Populations -- Euchromatic Variants -- Chromosome Heteromorphism (Summaries)- Fragile Sites -- Chromosome Variation Detected by Fluorescent In Situ Hybridization (FISH) -- Array-Comparative Genomic Hybridization/Microarray Analysis: Interpretation of Copy Number variants -- A CNV Catalog -- Gene and Genome Sequencing: Interpreting Genetic Variation at the Nucleotide Level
Abstract: This new edition now titled ́€œHuman Chromosome Variation: Heteromorphism, Polymorphism and Pathogenesiś€ provides the reader with an up-to-date overview of microarrays, fragile sites, copy number variations and whole genome sequencing. Greatly expanding the discussion of microarray analysis in the previous edition of the book, are new chapters on microarray and genomic analysis, plus comprehensive tables on the subtle microdeletions and microduplications that are found on each chromosome, including 235 recurring copy number variants that are associated with well-established or emerging chromosomal syndromes. The current edition features concise information on cytogenetic methods and applications, extending these discussions to DNA analysis and genome sequencing. Sections on euchromatin, heterochromatin, FISH pattern, fragile site, copy number, and DNA sequence variation are integrated with actual clinical examples from cytogenetic laboratories and from clinical practice. The principles that allow for the distinction between benign chromosome / DNA variation and pathogenic heteromorphisms / polymorphisms are discussed and include references to the latest organizational guidelines and genomic or population databases. The two previous incarnations of this book: the €˜Átlas of Human Chromosome Heteromorphisḿ€™, and ́€˜Human Chromosome Variation: Heteromorphism and Polymorphisḿ€™ have been standard reference works in most cytogenetic laboratories, used by laboratory directors and clinicians all around the world. While widely used sections from the previous edition on cytogenetic technologies and heteromorphisms are retained intact the present volume adds extensive material on copy number variations (polymorphisms detected by microarray analysis), fragile sites in disease and cancer, and practical views on interpreting emerging technologies, including whole exome sequencing. This book should be of interest to clinicians, technicians and students who are or will be exposed to DNA and/or chromosome analysis and the data derived from these continuously developing techniques. This fully updated book volume will bring the reader up to speed on the latest technologies, their applications, benefits and drawbacks and as such, is a must read for anyone with an interest in DNA and chromosome analysis and the distinction between benign variation and pathogenic mistakes

Abstract: Molecular Therapies of Cancer comprehensively covers the molecular mechanisms of anti-cancer drug actions in a comparably systematic fashion. While there is currently available a great deal of literature on anti-cancer drugs, books on the subject are often concoctions of invited review articles superficially connected to one another. There is a lack of comprehensive and systematic text on the topic of molecular therapies in cancer. A further deficit in the relevant literature is a progressive sub-specialization that typically limits textbooks on cancer drugs to cover either pharmacology or medicinal chemistry or signal transduction, rather than explaining molecular drug actions across all those areas; Molecular Therapies of Cancer fills this void. The book is divided into five sections: 1. Molecular Targeting of Cancer Cells; 2. Emerging and Alternative Treatment Modalities; 3. Molecular Targeting of Tumor-Host Interactions; 4. Anti-Cancer Drug Pharmacokinetics; and 5. Supportive Therapies
