Abstract: In the past decade, we have experienced an explosion of new information about cancer therapeutic targets. Many of the targets have been validated by the discovery and approval of new medicines which have been approved for the treatment of cancer. On the heels of these successes, innumerable new targets and new potential therapeutics are being developed by many different groups including government agencies, pharmaceutical companies, biotechnology companies, academic institutions, and individual investigators. Understanding the expanding "universe" of cancer therapies is therefore becoming impossible and no single source exists which serves as a reference for the involved parties. Further, the interested parties have vastly different areas of expertise, from focused laboratory based science, to clinical research, to corporate and regulatory oversight. The text would be updated every two years, more often depending on pace of change, interest and sales. While useful online, this reference book would likely be kept in hard copy as well

Abstract: This edited volume is about novel targeted therapies for the treatment of advanced neoplasms of the head and neck (HNSCC) with particular reference to oral squamous cell carcinomas and covers specific fields of cancer genetics and genomics for personalized medicine. The landmark studies from the HNSCC Cancer Genome Anatomy (TCGA) and the cancer Genome Atlas 2015 have revealed the multiplicity and diversity of genetic alterations in HNSCC. Abstract: This volume provides the reader with an overview of the diverse functions of the RUNX family of genes. As highlighted in the introduction and several of the 29 chapters, humans and other mammals have three RUNX genes that are known to play specific roles in blood, bone and neuronal development. However, their evolutionary history has recently been traced back to unicellular organisms and their involvement in many well-known signaling pathways (Wnt, TGFb, Notch, Hippo) is indicative of a more general function in cell biology. Their documented roles in cell fate decisions include control of proliferation, differentiation, survival, senescence and autophagy. The pleiotropic effects of RUNX in development are mirrored in cancer, where RUNX genes can function as oncogenes that collaborate strongly with Myc family oncogenes or as tumour suppressor genes. In the latter role, they display hallmarks of both ‘gatekeepers’ that modulate p53 responses and ‘caretakers’ that protect the genome from DNA damage. Several chapters focus on the importance of these genes in leukemia research, where RUNX1 and CBFB are frequently affected by chromosomal translocations that generate fusion oncoproteins, while recent studies suggest wider roles for RUNX modulation in solid cancers. Moreover, RUNX genes are intimately involved in the development and regulation of the immune system, while emerging evidence suggests a role in innate immunity to infectious agents, including HIV. At the biochemical level, the RUNX family can serve as activators or repressors of transcription and as stable mediators of epigenetic memory through mitosis. Not surprisingly, RUNX activity is controlled at multiple levels, this includes miRNAs and a plethora of post-translational modifications. Several chapters highlight the interplay between the three mammalian RUNX genes, where cross-talk and partial functional redundancies are evident. Finally, structural analysis of the RUNX/CBFB interaction has led to the development of small molecule inhibitors that provide exciting new tools to decipher the roles of RUNX in development and as targets for therapy. Abstract: Cancer is a multifaceted and genomically complex disease and data obtained through high throughput technologies has provided near complete resolution of the landscape of how genomic, genetic and epigenetic mutations in cancerous cells effectively influence homeostasis of signaling networks within these cells, between cancerous cells, tumor microenvironment and at the organ level. Abstract: This book is dedicated to the great scientist and outstanding individual Nikolay Wladimirovich Timofeeff-Ressovsky. The book brings together a number of brief stories/essays about Timofeeff-Ressovsky including “Stories told by himself”, and scientific chapters addressing his major research areas: genetics, radiobiology, radiation ecology and epidemiology, and evolution. Timofeeff-Ressovsky contributed to several fields of biology and established new directions of scientific research. He often repeated the phrase, which would later become famous: “Science should not be approached with the ferocity of wild animals”. In keeping with that philosophy, the issues discussed here are still open. Each scientific part starts with a current review; the chapters present leading scientific schools and views. The main theme discussed in the genetics part is mutation variability in the context of linear (replication, transcription, translation) and conformational template processes, and its dependence on phylogenetic group. In turn, the radiobiology chapters focus on the reorganization of DNA, cell, and population variability under low-dose irradiation, sparking indirect processes and adaptive response. The radiation ecology and epidemiology parts present data on the consequences of nuclear plants and related accidents for ecological systems and human beings. Here some approaches to estimating radiation risks are also offered. Evolution laws are demonstrated in the genomic universe, plant-microbe symbiosis, stabilizing and destabilizing (directional) selection. The last essay demonstrates the principles of organization operating in local animal populations, which are approached as social organisms of complex systemic nature.
